动物模型中进行的临床前实验证明免疫共刺激分子4-1BB/CD137的激动剂可以增强细胞毒T细胞和NK细胞的活性，诱导有力的抗肿瘤反应。Utomilumab（PF-05082566）是免疫共刺激分子4-1BB/CD137的激动性单抗。CLIN CANCER RES近期发表了一篇文章，评估Utomilumab治疗晚期恶性肿瘤的安全性、耐受性、药代动力学和药效学。

动物模型中进行的临床前实验证明免疫共刺激分子4-1BB/CD137的激动剂可以增强细胞毒T细胞和NK细胞的活性，诱导有力的抗肿瘤反应。Utomilumab（PF-05082566）是免疫共刺激分子4-1BB/CD137的激动性单抗。CLIN CANCER RES近期发表了一篇文章，评估Utomilumab治疗晚期恶性肿瘤的安全性、耐受性、药代动力学和药效学。

研究结果表明，Utomilumab的耐受性良好。在各个剂量水平均没有患者出现剂量限制毒性。最常见的治疗相关不良反应为疲劳，发热，食欲下降，头晕和皮疹。只有1例患者出现治疗相关3-4级不良反应（疲劳），未观察到临床相关的转氨酶升高。Utomilumab在0.006mg到10mg/kg剂量之间时表现出线性药代动力学特征。实体肿瘤的总客观有效率为3.8%，梅克尔细胞癌患者的总客观反应率为13.3%，包括1例完全缓解和1例部分缓解。循环生物学标志支持Utomilumab与4-1BB/CD137互相作用，提示循环淋巴细胞水平可能会影响临床获益。

文章最后认为，Utomilumab的良好安全性及初步的抗肿瘤活性值得进一步在恶性肿瘤患者中进行研究。
