Jennifer L. Gerton课题组近日发文,利用计算和微滴式数字PCR的方法,在人类和小鼠的正常细胞和癌细胞中测量核糖体DNA的拷贝数。他们发现,核糖体DNA的拷贝数和序列在癌细胞中会有所改变。人类癌细胞基因组内的拷贝数会有所减少,序列也更具多样化。具有低拷贝数的癌细胞基因组会出现mTOR的高活性。PTEN磷酸酶是一个对基因组稳定性起着重要作用的肿瘤抑制因子,也是mTOR激酶途径的负调节因子。在PTEN敲除的小鼠造血肿瘤干细胞中,核糖体DNA的拷贝数也比较低。同时,细胞的增值能力,核糖体DNA的产生水平和蛋白合成水平会提高。

核糖体DNA丢失一般发生在早期,并且和DNA对损伤的超敏感性相关。因此,核糖体DNA的拷贝丢失在癌细胞中是一个常见的现象,和 mTOR活化相关。

因此,核糖体DNA拷贝数可以作为一个简单的指标来评估癌细胞是否对DNA损伤的疗法敏感。
