研究表明ANGPTL3基因突变或抑制与人体主要脂质组分减少和动脉粥样硬化性心血管疾病发病率降低相关

血管生成素样蛋白3基因(ANGPTL3)功能丧失突变与血浆甘油三酯,低密度脂蛋白(LDL)胆固醇和高密度脂蛋白胆固醇(HDL)水平降低有关,但该突变或ANGPTL3抑制与动脉粥样硬化性心血管疾病的风险的关系尚不明确。同期还有一项研究表明该基因与血脂代谢有关:NEJM:Angptl3 反义寡核苷酸与血脂异常

在一项DiscovEHR人类遗传学研究中,研究人员对58335名受试者进行外显子测序,将ANGPTL3功能丧失突变与脂蛋白水平和冠心病发病相关联。同时在小鼠模型以及健康志愿者中测试单克隆抗体evinacumab对甘油三酯和低密度脂蛋白胆固醇的作用。

研究发现,ANGPTL3功能突变的杂合参与者其血清甘油三酯,高密度脂蛋白胆固醇和低密度脂蛋白胆固醇水平显著降低。有0.33%的ANGPTL3突变参与者患冠心病,而对照者患冠心病的比例为0.45%(调整后的比值比0.59,95% CI 0.41--0.85; P=0.004)。小鼠模型中,evinacumab抑制ANGPTL3可显著降低动脉粥样硬化病变面积和坏死含量。人体中,相比于安慰剂。evinacumab显示剂量依赖的甘油三酯和LDL胆固醇降低作用,最大降幅分别为76% 和23%。

研究表明ANGPTL3基因突变或抑制与人体主要脂质组分减少和动脉粥样硬化性心血管疾病发病率降低相关。
