加利福尼亚大学研究人员的一项研究显示,新研发的阶梯式过程能将小鼠皮肤细胞转化为分泌胰岛素的β细胞。Hampton博士对此进行了报道,文章发表在2014年4月2日的The Journal of American Medical Association上。【原文下载】

当研究人员采用独特的分子和重组因子混合物处理成纤维细胞时,这些细胞转化为内胚层样细胞。进一步用其它化合物处理,细胞转化为胰腺祖细胞样细胞(PPLCs),进而可被诱导为成熟的胰岛素分泌细胞。

将PPLCs移植到高血糖小鼠体内1周后,动物血糖水平逐渐降低。移植8周以后,PPLCs产生具有完全功能性的胰岛β细胞。

由于胰岛β细胞再生能力有限,难以大量生成。这一最新的过程使细胞生命周期延长。除了增加被1型糖尿病破坏的胰岛素分泌细胞,该研究成果使人们对导致疾病的β细胞先天性缺陷有了进一步的认识。
