STTT:中山医学院鉴定出M1病毒的特异性受体!奠定OVM精准疗法的科学基础

该研究将推动M1病毒的精准诊断和临床转化,并为溶瘤病毒疗法的生物标志物研究提供范式。

溶瘤病毒疗法是一种新型的恶性肿瘤免疫治疗策略,具有高靶向性、多机制溶瘤和可编辑改造等优势,正在逐渐成为肿瘤免疫治疗的热点领域。尽管溶瘤病毒在部分难治性恶性肿瘤病人上取得了惊人的疗效,仍有很大比例的病人无法从中获益,其根本原因在于缺乏用于区分响应患者和非响应患者的精准诊断生物标志物。根据病毒的生活史特点,溶瘤病毒对肿瘤的杀灭取决于两个关键步骤:通过胞膜受体进入肿瘤细胞和在胞内成功完成基因组复制,这提示溶瘤病毒存在胞膜型和胞内型两大类标志物。我院颜光美教授团队于2014年将M1病毒原创性地鉴定为一种新型溶瘤病毒(PNAS, 2014)后,陆续发现了四个预判M1病毒疗效的胞内型生物标志物,分别是锌指抗病毒蛋白ZAP(Oncogene, 2020)、胆固醇通路相关蛋白RHOQ(Nature Communications, 2018)、内质网应激通路蛋白IRE1α(Journal of Virology, 2018)和KRAS突变(Molecular Oncology, 2020)。

2022年4月8日,我院颜光美教授团队在重要知名科学期刊Signal transduction and targeted therapy(STTT)杂志在线发表题为 “Identification of the receptor of oncolytic virus M1 as a therapeuticpredictor for multiple solid tumors” 的Article研究论文。该研究鉴定了MXRA8为新型溶瘤病毒M1(OVM)的受体,原创性地确定了预测M1病毒疗效的双生物标志物(dual-biomarker)体系。

在本项研究中,研究团队首先利用多细胞转录组学进行Weighted Gene Co-Expression Network Analysis(WGCNA)分析,筛选出膜蛋白MXRA8是介导M1病毒杀伤肿瘤细胞的关键宿主因子;随后的细胞水平和动物水平过表达和敲除实验证实MXRA8正向调控M1病毒的溶瘤效应;进一步地,研究人员使用冷冻电镜技术获得了MXRA8与M1病毒相结合的低分辨率结构,直观地证明MXRA8是M1病毒的受体。同时,研究人员发现MXRA8在实体瘤中广泛高表达,其表达量显著高于癌旁配对正常组织。以上结果提示受体MXRA8可作为M1病毒的胞膜型生物标志物。重要的是,研究人员发现在肿瘤细胞株和病人肿瘤组织中,ZAP的低表达和MXRA8的高表达能共同预测M1病毒的组织活性抑制能力。综上所述,该研究鉴定了新型溶瘤病毒M1的受体,并提出使用胞膜受体MXRA8和宿主因子ZAP做为预测M1病毒疗效的双生物标志物(dual-biomarker)体系。该研究将推动M1病毒的精准诊断和临床转化,并为溶瘤病毒疗法的生物标志物研究提供范式。
