由于结核分枝杆菌耐药性复杂，需要进行快速、全面的药敏试验来指导患者的治疗。本研究开发了基于Illumina MiSeq和Nanopore MinION的靶向测序工作流程，用于预测结核分枝杆菌对12种抗生素的耐药性。

从香港和埃塞俄比亚收集的163m结核分离株进行了多重PCR，同时扩增了19个耐药相关基因区域。然后分别在MiSeq和MinION上对24个和12个样品进行条形码和测序。采用一个基于网络的生物信息学流程：BacterioChek-TB用来将原始数据集转化为对临床人员有利的报告。

研究结果显示，两个平台均成功测序了样品的平均读数深度分别为1127和1649倍。如果排除MinION报告的等位基因频率<40%的变体，MiSeq和MinION的变体信号一致可以达到100%。与表型药敏试验(pDST)相比，两种工作流程的平均临床敏感性为94.8%，临床特异性为98.0%。MiSeq和MinION工作流程的周转时间分别为38和15小时，分别比pDST提前17-18天提供治疗指导。MinION平台(71.56美元)和MiSeq平台(67.83美元)上的每个样本成本较高，这是因为批处理能力不同。

研究证明了MiSeq和MinION平台的互换性，为结核病的治疗产生准确和可操作的结果。
