研究发现抗炎药具有癌症治疗前景

塞来昔布是常用的非甾体类抗炎药,作用靶点为环氧化酶-2(COX-2),COX-2是一种与体内炎症相关的酶类,此外,该酶在促进肿瘤的生长过程中也起着关键作用。COX-2在正常组织中活性很低,但在多种类型肿瘤中却很高。

专家们感兴趣的是哪些特定信号转导通路确实对癌症进展产生了影响。在研究过程中,他们发现,癌症中一些信号能够激活炎症相关的酶类,包括COX-2,然而像塞来昔布一类的抗炎药能够阻止这一过程。

接着,研究者继续进行动物实验以探索塞来昔布对II型多发性神经纤维瘤细胞(NF2)生长的抑制作用。在人类当中,由于抗肿瘤基因的突变可引起NF2的出现,这将引起听觉神经出现良性肿瘤。结果发现,每天接受塞来昔布处理的动物其肿瘤生长率显着低于模型组。

进一步研究发现,Hippo-YAP通路在此过程中起着重要作用,YAP是NF2细胞生长和分裂所必需的,并且与肿瘤形成有关。研究者称本次的研究显示COX-2抑制剂对抑制肿瘤细胞生长有作用,他们相信,COX-2抑制剂对其他与炎症相关的癌症也有类似作用。
