机体如何及时启动天然免疫反应以有效地清除病原体入侵，同时又适度控制免疫应答强度并能够及时终止免疫应答而不损伤机体自身组织？这是免疫学根本性关键科学问题，对于免疫系统功能的发挥与机体自身稳定至关重要。12月29日出版的新一期《自然免疫学综述》（Nature Reviews Immunology）发表了中国工程院院士、中国医学科学院院长曹雪涛的长达16页的综述性评论文章，系统总结了天然免疫应答领域的研究成果，分析了该领域存在的挑战性问题，归纳性地提出了天然免疫应答5种调控模式并前瞻性地提出了11个重要研究方向。

近20年来，免疫学家对于天然免疫识别、应答与调控的细胞与分子机制取得了许多进展，对于炎症的分子机制与调控紊乱所导致的众多疾病也有了更深入的认识，提出了许多新的干预靶点以及新的治疗策略。2011年诺贝尔生理或医学奖授予了天然免疫应答机制反映出了该领域的重要性和前沿性。如何总结以往工作并在此基础上寻找新的突破性研究方向，是免疫学界需要重新思考的重要问题。

在天然免疫与炎症的研究领域，模式识别受体（PRR）的结构与功能研究是该领域核心研究内容与热点，曹雪涛院士将这些研究结果进行了系统性总结，从天然免疫应答自身调控的角度，提出了5种调控模式，包括PRR蛋白降解、亚细胞转位、诱导性表达、募集抑制因子与辅助增强子模式；从天然免疫应答交叉调控的角度，提出了5种调控模式，包括协同效应、增强效应、阻抑效应、反馈抑制效应和反馈增强效应。在此基础上，提出了该研究领域值得关注的11个研究方向。

据悉，国际顶级学术杂志会定向邀请某个领域有学术影响力并对于该领域有实质性贡献的资深著名科学家就特定研究领域进行回顾性总结，并根据自身研究的体会与思考，站在学科前沿提出该领域面临的挑战性难题与展望未来研究方向。这是我国学者第一次应邀为《自然免疫学综述》撰写综述性评论长文，也客观反映出我国免疫学家在国际同行中已经具有相当的学术影响力。
