Stem Cell Rep:电流引导神经干细胞体内迁移

神经干细胞的移植具有治疗多种神经疾病的潜力。研究人员已经证明,神经干细胞能够迁移并且分化形成大鼠的脑部和脊髓。但是神经干细胞在人类大脑中的迁移却比较困难,因为人脑的体积较大,神经干细胞需要迁移的路程较长。此外,移植的神经干细胞的迁移能力一般比较差,因为移植神经干细胞之间,及其对子代细胞,具有抑制迁移的现象。因此,提高神经干细胞在脑部的迁移能力有助于神经干细胞在人类疾病中的应用。

加州大学戴维斯分校的研究人员报导了一个新的促进和引导神经干细胞在体迁移的方法。研究人员开发了一种安全的刺激手段,在大脑中传递方向性的电流。利用GFP的标记,研究人员得以观察神经干细胞的在体迁移。他们发现,电流能够诱导神经干细胞的迁移,甚至能够指导人类神经干细胞逆着内在的迁移信号移动。电流刺激之后3周,研究人员在目标区域观察到了迁移而来的移植神经干细胞。在刺激之后四个月,在目标区域观察到了移植神经干细胞的分化。

因此,利用电流引导神经干细胞的迁移可能可以促进干细胞在脑部的应用。
