Apoptosis:七叶皂苷在乳腺癌治疗中的作用——诱导铁死亡的潜在机制及与顺铂的协同抗肿瘤活性

探索了铁死亡和G6PD在七叶皂苷(Escin)诱导的乳腺癌(BC)细胞死亡中的作用及其潜在机制,并探讨了Escin和顺铂之间的协同作用。

背景:乳腺癌(BC)长期以来一直威胁着世界各地的女性,需要新的治疗方法。铁死亡是一种新型的调节性细胞死亡,是BC的潜在治疗靶点。该研究探索了铁死亡和G6PD七叶皂苷(Escin)诱导的BC细胞死亡中的作用及其潜在机制,并探讨了Escin和顺铂之间的协同作用

方法:CCK8法测定人BC细胞系(MDA-MB-231和MCF7)的生长情况;流式细胞术检测细胞凋亡率和ROS含量;试剂盒检测GSH、G6PD和脂质过氧化水平;qRT‒PCR、WB、免疫荧光和免疫组化检测相关蛋白表达;siRNA转染和病毒感染G6PD或GPX4进行验证。

将稳定表达sh-NC和sh-G6PD的MDA-MB-231细胞注射到4周龄雌性BALB/c裸鼠右髂窝(每只小鼠2.5×106个细胞)。当肿瘤生长到大约60 mm3时,用生理盐水作为对照腹膜内注射2mg/kg的Escin。当肿瘤体积达到约300mm3时,通过暴露于CO2对裸鼠实施安乐死,并取出结节进行分析。

结果:Escin在体外和体内抑制BC细胞生长,铁死亡可能是Escin诱导的细胞死亡的主要原因。从机制上讲,Escin显著下调GPX4的蛋白水平,而GPX4过表达可以逆转Escin引发的铁死亡。进一步的研究表明,Escin可以促进G6PD的泛素化和降解,从而抑制GPX4的表达,并有助于铁死亡。

此外,蛋白酶体抑制剂MG132或G6PD过表达可以部分逆转Escin诱导的铁死亡,而G6PD敲低会加重这种情况。体内研究还支持G6PD的下调加剧了Escin对肿瘤生长的抑制。最后,在BC细胞中,Escin与顺铂联合可显著提高细胞凋亡。

结论:Escin通过调节G6PD/GPX4轴介导的铁死亡,在体内和体外抑制肿瘤生长。该研究发现为BC提供了一种有前景的治疗策略。
