腹膜内注射一克他croline(60 mg/kg)诱发肺动脉高压。从单crocrotaline注射后的第七天开始,每天腹膜内施用MgSO4(100 mg/kg),持续3周。将成年雄性大鼠随机分为单丁香碱(MCT)或单丁香碱加MgSO4(MM)组(每组n = 15);每组15只。对照组同时维持。为了进行分析,将存活的大鼠在接受克他crotaline后第28天处以安乐死。

 

结果显示,MM组的生存率高于MCT组(100% vs 73.3%,p = 0.043)。MM组的肺动脉壁增厚,α-平滑肌肌动蛋白上调,右心室收缩压升高和右心室肥大水平均低于MCT组(所有p <0.05)。MM组的脂质过氧化、线粒体损伤、炎性体和细胞因子上调水平以及肺和右心室的凋亡水平均低于MCT组(所有p <0.05)。值得注意的是,外源氯化钙抵消了MgSO4对肺动脉壁增厚和右心室肥大的缓解作用。

 

总之,该研究结果表明,MgSO4可能通过拮抗钙来减轻肺动脉高压的发展。
