除了积极地将药物运出细胞外,Pgp还介导癌细胞中化学治疗药物的溶酶体螯合作用,从而有助于耐药性的产生。
最近,来自德国的研究人员发现,包括多柔比星在内的Pgp底物的溶酶体螯合作用也发生在形成血脑屏障的人和猪脑内皮细胞中。接下来是封存药物的囊泡结构的脱落,其保持附着于质膜的顶侧,并形成最终经历嗜中性粒细胞吞噬作用的聚集体(“屏障体”),构成了尚未描述的药物排出机制。
这些发现引入了一种机制,可能有助于脑保护,防止潜在的毒性外来分子,包括治疗上重要的化疗药物。
