Science Advances:北理工黄渊余团队开发新型脂质纳米颗粒,递送RNAi疗法,治疗和预防心血管疾病

高血脂症或心血管疾病是世界范围内主要的死亡原因,严重威胁人类的生命健康。除了建立健康的生活方式外,服用药物是控制疾病的重要方法。目前,降脂治疗干预旨在降低低密度脂蛋白胆固醇(LDLc),即所谓的“坏胆固醇”,而对甘油三脂(TG)几乎没有影响。然而,甘油三酯与冠状动脉疾病息息相关。因此,开发可同时降低低密度脂蛋白胆固醇(LDLc)和甘油三脂(TG)的治疗药物对于治疗高脂血症和预防心血管疾病具有重要意义。

目前,抗高血脂药物主要包括小分子药和抗体药,然而他们在使用过程中存在潜在的有害影响,且治疗效率低,严重阻碍了治疗进展。幸运的是,小干扰RNA(siRNA)由于具有开发时间短、特异性高、高度可预测等优势成为下一代降脂药物开发的关键。脂质纳米颗粒(LNPs)已经被广泛应用于siRNA递送,可显着改善siRNA的药代动力学和药效学特征,并成功推动了RNAi疗法首次获得FDA批准。因此,开发和优化具有高运输效率和理想体内安全性的新型LNP递送系统成为核心任务。

2022年2月16日,北京理工大学黄渊余教授团队在 Science 子刊 Science Advances 上发表了题为:Thermostableionizable lipid-like nanoparticle (iLAND) for RNAi treatment of hyperlipidemia 的研究论文。

该研究团队开发了一种新型热稳定可离子化脂质辅助核酸递送系统——iLAND,该系统具有良好的理化性质、热稳定性和优异的siRNA转运效率。

研究团队进一步验证了siRNA@iLAND在高脂饮食(HFD)喂养小鼠、db/db小鼠和hApoC3-Tg小鼠等三种疾病模型中的治疗效果,研究表明低剂量的siRNA即可达到了预期的治疗效果。

这项研究表明,iLAND可作为一种有前景的核酸递送平台,而siRNA@iLAND有望成为治疗高脂血症和预防后续代谢疾病的潜在治疗方法

该团队首先对iLAND的物理化学性质以及对siRNA的递送能力进行研究,研究表明,iLAND的流体动力学尺寸、PDI和zeta电位分别约为140nm、0.15和20mV,且具有脂质双层结构。iLAND具有优异的封装效率和负载效率,分别为90%和65%。iLAND在体外显示出高细胞摄取、内体逃逸效率和生物相容性,可与广泛使用的体外转染剂Lipo2000相媲美。此外,siRNA@iLAND具有优异的稳定性,在40℃、室温以及-20℃的条件下储存一周,未发生降解。

接下来,探讨了iLAND的内化和细胞内贩运过程行为和机制,研究表明siRNA@iLAND主要由小窝依赖性内吞作用介导。载脂蛋白E(ApoE)在介导细胞摄取方面发挥了关键作用。内化后,iLAND进入内体后通过内体膜不稳定机制、质子海绵效应和渗透膨胀作用,触发siRNA从内体(或溶酶体)快速有效的逃逸。此外,生物分布测定表明,siRNA@iLAND在肝脏中迅速且显着积累。

接下来,在三种高血脂症小鼠模型中进行了功效研究,包括HFD小鼠、db/db小鼠和hApoC3-Tg小鼠。选择了两种siRNA,其中siANG应用于前两个模型,siApoC3应用于hApoC3-Tg小鼠。研究表明,与正常小鼠相比,siRNA@iLAND作用于三种高血脂模型后,不仅可以下调血清胆固醇水平,还可以降低血清甘油三酯水平,对治疗高血脂症和预防心血管疾病非常有益。

总体而言,iLAND具有优异的siRNA转运效率,且siRNA@iLAND在高脂血症治疗和预防后续代谢疾病方面具有重要价值,有望为高血脂症患者带来福音
