卵巢透明细胞癌mTORC1/2治疗策略

晚期卵巢透明细胞癌（OCCC）对常规的铂类化疗无反应。OCCC中常见的改变包括肿瘤抑制因素ARID1A中的有害性突变以及PIK3CA激活性突变。CLIN CANCER RES近期发表了一篇文章，寻找OCCC患者中未知的突变酶并在OCCC模型中检测下游通路作为药物靶点的可行性。

晚期卵巢透明细胞癌（OCCC）对常规的铂类化疗无反应。OCCC中常见的改变包括肿瘤抑制因素ARID1A中的有害性突变以及PIK3CA激活性突变。CLIN CANCER RES近期发表了一篇文章，寻找OCCC患者中未知的突变酶并在OCCC模型中检测下游通路作为药物靶点的可行性。

作者在OCCC患者队列中进行测序，检测人激酶组，肿瘤相关基因以及拷贝数变化。使用OCCC细胞系及异种移植模型进行ERBB TKI类药物erlotinib和lapatinib，PARP抑制剂olaparib和mTORC1/2抑制剂AZD8055药物测试。研究结果表明，在OCCC中发现了几种之前注意到的低频率突变。结合突变和拷贝数改变，91%的肿瘤涉及到PI3K/AKT/mTOR通路，MAPK通路或ERBB家族。82%涉及到DNA修复通路。OCCC患者中强p-S6染色表明高mTORC1/2活动度。主要的OCCC细胞系对AZD8055阻断mTORC1/2敏感，对靶向ERBB家族或DNA修复信号的药物不敏感。在异种移植模型中进一步验证了mTORC1/2抑制的有效性。

文章最后认为，mTORC1/2抑制是OCCC有效的治疗策略。
