Science:如何寻找对有益的基因突变?

对于很多人来说,基因突变并不是褒义词,但并不是所有的基因突变都那么坏。科学家表示,有些基因突变甚至能使机体保持健康,可以抵消或抑制其他突变带来的坏影响。近日,科学家们在Science杂志上揭示了如何寻找到这些有益的突变,对了解基因抑制过程又重要的意义。人们通常认为基因突变给机体带来了疾病,但并不是所有的突变都是罪魁祸首,有些甚至可以抵消或抑制某些突变带来的恶性结果,该过程被称为基因抑制,且鲜为人知

人们通常认为基因突变给机体带来了疾病,但并不是所有的突变都是罪魁祸首,有些甚至可以抵消或抑制某些突变带来的恶性结果,该过程被称为基因抑制,且鲜为人知。11月4日,多伦多大学的研究人员揭露了该过程的基本原理,这为人们了解基因抑制过程奠定了基础。

有些突变能抵消有害突变带来的恶性结果

该研究由Donnelly中心及分子遗传学系的Brenda Andrews教授、 Charles Boone 教授、Frederick Roth教授以及明尼苏达大学双城分校Chad Myers教授引领,他们编译了细胞中的首个抑制突变综合集,相关结果发表在science杂志中。这些结果有助于解释抑制性突变如何与致病突变相结合,从而减轻疾病症状,甚至能完全抑制致病突变的作用。

“同样是携带损害性突变,为何有些人会生病而有些却完全健康?其部分原因可能与环境有关,但大部分原因是因为其他突变的存在,这些突变抑制了第一个突变的效应。”本文资深作者Roth教授说。

打个比方说,当被困在恒温器坏掉了的房间时,随着时间推移,房间温度越来越高,这种情况要把温度降下来,你可以修复温度调节器,或者打破一个窗口,基因抑制过程保持细胞健康的原理与之类似。

Science上的这项研究开辟了人们对基因抑制过程的新理解,甚至会带来新的遗传性疾病治疗方法。“如果我们知道哪些是抑制基因,那么就可以了解它们是如何与疾病基因相互作用,这有助于指导未来的药物开发。”研究人员表示。

解开基因抑制的基本原理

然而,抑制性突变是很难被发现的,寻找它们就好比大海捞针。理论上,DNA中成千上万个突变中的任何一个都有可能是抑制性突变,更何况人类有那么多个基因,因此要发现所有的抑制性突变是不切实际的。不过,通过这项研究,将来寻找抑制性突变也许就不再是大海捞针。

研究人员表示,寻找抑制基因时,我们无需把范围放到离损害性突变基因很远的位置,因为它们在细胞中有类似的作用——所编码的蛋白质可能错落在同一个位置,或者分享了同一个分子途径。“我们已经发现了基因抑制作用的基本原理,揭示了有害突变基因的抑制基因通常是功能与其相关的基因。寻找抑制基因并不是大海捞针,例如寻找人类基因疾病的抑制基因时,我们可以缩小范围,将20000个基因缩小到数百个,甚至数十个,这是一个很大的进步。”

“像这样的研究,在全球范围内从未开展过,因为在人类身上做实验室是不可能的,因此我们使用了酵母作为模型生物,通过酵母我们可以了解突变如何影响细胞的健康。” Van Leeuwen说。酵母细胞包含了6000个基因,是人类细胞的简单版本,其遗传学的基本原则也适用于人类。此外,去除酵母细胞中的任何一个基因也相对简单,这有助于研究突变基因对细胞的影响。

该研究团队采用了双管齐下的方法,一方面,他们分析了已报道的所有酵母基因之间的抑制关系,而原有的这些数据不可避免地偏向于一些已被科学家们报道的较流行的基因。另一方面,研究人员还进行了无偏向分析,检测当细胞携带损害性突变时该如何生长,或如何与其他突变相互作用。由于有害突变减缓了细胞的生长,因而任何生长的改善都有可能受益于第二个基因的抑制性突变。该研究揭示了与已知损害性突变相互作用的数百个抑制性突变。
