SCI REP:成纤维细胞生长因子23减弱微流体装置中人血液中性粒细胞的趋化性

嗜中性粒细胞在组织中的运输,对调节身体的免疫反应有重要影响。嗜中性粒细胞迁移可以在宿主防御中起保护作用,同时也可以引起健康问题。成纤维细胞生长因子23(FGF23)是已知的慢性肾脏疾病(CKD)生物标志物,最近研究显示,成纤维细胞生长因子23也会损害中性粒细胞在内皮的滞留以及跨内皮的迁移。

嗜中性粒细胞在组织中的运输,对调节身体的免疫反应有重要影响。嗜中性粒细胞迁移可以在宿主防御中起保护作用,同时也可以引起健康问题。成纤维细胞生长因子23(FGF23)是已知的慢性肾脏疾病(CKD)生物标志物,最近研究显示,成纤维细胞生长因子23也会损害中性粒细胞在内皮的滞留以及跨内皮的迁移。在本研究中,我们使用两个新的微流体装置来进一步检测FGF23对人血液嗜中性粒细胞趋化性的影响。我们的研究结果表明,无论在FGF23存在或不存在的背景下,FGF23预处理的嗜中性粒对照细胞相比,在fMLP梯度的趋化性试验中,fMLP的浓度均较低。同时,FGF23预处理的细胞流动性更强。FGF23对嗜中性粒细胞迁移的影响以及和多细胞迁移参数之间的相关性,被进一步通过趋化熵和主成分分析揭示。总的来说,这些结果揭示了FGF23对减弱嗜中性粒细胞趋化性的作用,并且揭示了FGF23介导的嗜中性粒细胞迁移与一些疾病如CKD的直接相关性。
