CLIN CHEM:液体活检靶向重测序如何多重消除野生型DNA?

从液体活组织检查中收集的临床样品和循环的无细胞DNA(cfDNA)在癌症中的诊断和预后的应用正在迅速发展,人们希望一种能够有减少过量野生型(WT)样品部分影响的改良方法。近日,国际杂志 《CLIN CHEM》在线发表一项关于使用WT 的DNA酶降解来对包含突变序列进行突变富集,突变富集与在多重闭管反应中进行的高分辨率熔解(HRM)结合,作为靶向重测序之前的快速有效的筛选工具。 研究人员开发了一种均匀的闭管方法,该方法使用双链DNA特异性核酸酶同时在多个靶标上降解WT DNA。采用探针重叠(ND-NaME-PrO)的无变性核酸酶辅助的次要等位基因富集利用WT寡核苷酸在推定的DNA靶标上将两条链重叠。在部分变性(DNA呼吸)的条件下,寡核苷酸探针在选定的靶标上增强双链DNA特异性核酸酶消化,从而使对WT偏好高于突变体DNA。为了验证ND-NaME-PrO,研究人员使用多重HRM,数字PCR和MiSeq靶向重测序突变基因组DNA和cfDNA。 研究显示含有KRAS突变的DNA的连续稀释显示突变富集为10至120倍,等位基因检测可低至0.01%。复制的ND-NaME-PrO与多重PCR-HRM
