自人类对疾病认识进入后基因组时代以来,科学家一直试图测定病毒的内部三维结构,这一工作近日被两位中国科学家率先完成。

9月18日,国际顶级学术期刊《Science》杂志发表了一篇题为“冷冻电镜揭示双链RNA病毒内部聚合酶和病毒基因组的非线轴结构”的论文。论文中,两位中国科学家首次将病毒的观察视角从“衣壳”深入到内部,改变了过去认为该类型病毒的内部基因组应呈线轴状排列的主流观点,亦用一种全新方法,开启帮助人类认知更多生物分子内部结构的大门。

这两位科学家分别是湖南师范大学物理与信息科学学院教授、清华大学生命科学学院副研究员程凌鹏。刊登在《Science》杂志的论文也是他们合作研究完成的。

近年来,结构生物学家利用电子显微技术与X射线晶体学技术,已解析大量病毒衣壳的近原子分辨率三维结构,对衣壳蛋白分子结构形成了较为系统的认知,但对病毒内部的基因组及相关蛋白三维结构,迄今依旧一无所知。

经刘红荣、程凌鹏两位科学家在病毒三维重构领域的10余年合作研究,他们最近提出了一种基于二维电子显微图像解析对称失配生物大分子三维结构的新方法,并运用这个方法首次解析一种源自昆虫的双链RNA病毒内部基因组,以及其RNA聚合酶的三维结构,发现该病毒内部结构呈现“多层球状”。

刘红荣表示,他们此番取得的研究成果,首次求解了衣壳内部基因组及相关蛋白三维结构,向病毒学领域证明了衣壳内部的核酸与蛋白质结构是可解的,势必推进人类重新认知病毒。

“我们这个研究更多的还是方法学,属于基础研究,对人类疾病防控及药物设计存在潜在的应用前景。”刘红荣透露,他们计划把取得成果的研究方法推广到其他的领域,尤其是生物大分子动态的三维结构研究。

据悉,刘红荣于2011年从美国加州大学洛杉矶分校博士后出站归国后,赴湖南师范大学工作,其团队主要负责从事冷冻电镜、凝聚态物理及其交叉科学的研究工作,近年来获得国家自然科学基金等项目的资助,并在国内外一流学术期刊发表多篇学术论文。
