最近,来自霍普金斯医学院的研究人员开发了一种笼式RNA策略,其允许Cas9与DNA结合,但在光诱导激活之前不会裂解。这种方法被称为非常快速的CRISPR技术(vfCRISPR),其在亚微米和第二尺度创建双链断裂(DSBs)。同步裂解改善了DNA修复的动力学分析,揭示了细胞在几分钟内对Cas9诱导的DSB作出反应,并可以在DNA连接后保留MRE11。

DNA损伤后H2AX的磷酸化每分钟传播超过10万碱基,达到3000万碱基。

利用单细胞荧光成像,研究人员表征了53BP1修复灶形成和溶解的多个周期,第一个周期比随后的周期时间长,其持续时间受修复抑制的调节。成像引导的亚细胞Cas9激活进一步促进了单个等位基因分辨率的基因组操作,

总之,vfCRISPR使DNA修复研究在空间、时间和基因组坐标上实现了高分辨率。
