糖尿病参与多种疾病的发病及其导致的死亡，包括心血管疾病和慢性肾脏疾病。随着肥胖人群的日益增加，2型糖尿病（T2D）正在成为一个世界性的难题。现今，有将近10%的成年人受其影响，每年有超过150万的死亡病例。肥胖引起的糖尿病的特征在于高血糖，胰岛素抵抗和进行性β细胞衰竭。

糖尿病参与多种疾病的发病及其导致的死亡，包括心血管疾病和慢性肾脏疾病。随着肥胖人群的日益增加，2型糖尿病（T2D）正在成为一个世界性的难题。现今，有将近10%的成年人受其影响，每年有超过150万的死亡病例。肥胖引起的糖尿病的特征在于高血糖，胰岛素抵抗和进行性β细胞衰竭。 在具有肥胖性糖尿病的小鼠胰岛中，来自华盛顿大学医学院的研究人员观察到β细胞死亡的增加和自噬流的受损。研究人员提出假设，临床上的可持续治疗手段，间歇性禁食，是通过促进自噬流，改善肥胖引起的糖尿病症状。研究人员的数据显示，在有持续性高脂肪摄入的情况下，间歇性禁食依旧可以恢复胰岛的自噬流，并通过增加葡萄糖刺激的胰岛素分泌，β细胞的存活，胰腺再生标志物NEUROG3的核表达，来提高葡萄糖耐受度。 相反的，在缺乏溶酶体膜蛋白LAMP2或是自噬关键基因BECN1 / Beclin-1单倍体不足的肥胖小鼠中，溶酶体的功能会出现障碍，间歇性禁食不能挽救β细胞的死亡或诱导NEUROG3的表达。此外，在LAMP2缺失的非肥胖性小鼠中，间歇性禁食足以引起β细胞死亡，证明了在饥饿条件下溶酶体功能对于维持β细胞体内平衡的重要性。在LAMP2-或
