可溶性环氧水解酶抑制剂

可溶性环氧化物水解酶（sEH）是花生四烯酸级联中的一种酶，其将环氧脂肪酸（EpFAs）（例如由细胞色素P450酶产生的环氧二十碳三烯酸（EET））转化为二羟基 - 二十碳三烯酸。在过去的20年中，随着sEH抑制剂的发展，在体内模型中提高EET和其他EpFAs的水平已经成为可能。最近有研究表明，EET在骨吸收过程中抑制炎症方面起着关键作用，但其机制尚不清楚。在目前的研究中，我们使用sEH抑制剂（1-三氟甲氧基苯基-3-（1-丙酰基哌啶-4-基）脲[TPPU]）来研究小鼠牙周炎模型中的免疫调节作用。

方法：在第0天，第2天和第4天时，用伴放线放线杆菌感染小鼠，并将其分组（n = 6），每天口服1mg / kg TPPU治疗，共15天。然后，处死小鼠并使用形态测定法分析它们的颌骨的骨吸收。通过微阵列PCR或蛋白质印迹分析牙龈组织中的免疫炎性标记物。

结果：用TPPU处理的感染小鼠比未经处理的感染小鼠骨吸收更低。并且，感染小鼠sEH的表达增加；然而，TPPU治疗的小鼠sEH表达降低。此外，在用1mg / kg TPPU处理的组中，几种促炎细胞因子和分子标记物在牙龈组织中下调。

sEH抑制剂TPPU在骨吸收小鼠模型中具有免疫调节作用，可以抑制骨吸收和炎症反应。
