Sci Tran Med:突破!纳米药物可治疗心脏疾病

近年来,纳米颗粒已被用于载药系统,负责将药物输送到身体的特定部位。这种疗法通常是口服或静脉注射,但往往心脏药物输送效率低下。为此,研究人员开发出了一种可吸入式纳米药物,这使得它们能够更快地到达心脏并被心肌细胞吸收,最终改善心脏功能。

这种新型可吸入式纳米药物由意大利和德国的研究人员共同研发,用于治疗心脏病患者。详细描述吸入剂发展、使用方法以及动物实验测试效果的相关研究结果发表在《Science Translational Medicine》上。

研究小组是使用与牙齿和骨骼相似的材料——“磷酸钙颗粒”制造出纳米颗粒的,它小到足以被心脏组织吸收,大到足以将药物运输到指定地方。该研究选择了可以修复心脏细胞表面的钙离子通道的药物,而修复钙离子通道被认为是恢复正常心电活动最关键的部分。

在药物负载在纳米颗粒上之后,研究小组以模仿糖尿病性心肌病的方式将其给予心脏受损的小鼠和大鼠,再通过观察从左心室排出多少血来测量心脏的健康状况。结果发现,在给药之前,心脏受损小鼠比健康小鼠心脏评分低17%。经过治疗,测量值平均上升了15%,而这是一个接近完全恢复的状态。

研究人员对啮齿类动物身上的研究结果很满意,随后还测试了猪的呼吸系统(其呼吸系统与人类更为相似),想知道药物在心脏组织中的富集速度。结果发现它的速度和预期一样快,相比传统方法也有改进。

研究小组还报告说,吸入纳米颗粒不会对小鼠或大鼠的心脏组织产生任何毒性。当然在人体内进行测试之前,还需要更多的研究来确保输送系统是安全的。
