正常发育期间的组织特异性转录过程需要其远端顺式调节元件(例如增强子)的严格控制。增强子是具有由转录因子、共激活因子和染色质重塑酶识别的特定DNA序列。Gata3是序列特异性DNA结合转录因子,其调节多种组织和器官的形成,包括内耳、晶状体、乳腺、T细胞、泌尿生殖系统和甲状腺等。在眼睛中,Gata3在晶状体囊泡的后部具有高度特异的表达域,然而,其潜在的调控机制尚不清楚。

在这里,作者鉴定出位于其转录起始位点上游约18kb的新型Gata3晶状体特异性增强子。他们发现,在晶状体中,5kb处的 Gata3启动子具有低活性。该增强子含有AP-1、Ets-、Smad1 / 5结合位点,以及与晶状体相关的DNA结合因子结合位点。瞬时转染研究显示,该增强子可以提高BMP4和FGF2的启动子活性。

因此,他们认为,这些研究鉴定了具有高活性的Gata3的新型远端增强子,并且表明BMP和FGF信号传导可通过该增强子上调启动子活性,进而促进Gata3在晶状体纤维细胞中的表达。
