Immunity:中国学者同期4篇论文,聚焦免疫进展

蒋争凡教授发现炎症小体激活caspase-1介导cGAS切割调控DNA病毒感染免疫反应。病毒感染触发宿主先天免疫应答,导致产生各种细胞因子,包括I型干扰素(IFN),炎症细胞的激活和受感染细胞的程序性细胞死亡。炎症细胞因子产生对于触发有效的免疫应答是关键的,所述有效的免疫应答应该是治疗感染而不引起宿主病理。在Asc -/- 和Casp1 -/-缺陷的巨噬细胞中,导致DNA病毒感染后,干扰素产生增多,但在RNA病毒病毒感染时并没有变化。这项研究表明,经典和非经典炎症小体激活后,caspase-1与cGAS相互作用,切割它并抑制cGAS-STING介导的IFN产生。炎症小体信号缺陷在体外和体内增强了对DNA病毒的宿主抗性,并且这种调节作用延伸到其它炎性半胱天冬酶。因此,炎症小体激活抑制cGAS依赖性信号,暗示细胞内DNA感测途径之间的交叉调节。

施福东发现脑缺血时抑制免疫功能的机制。在脑缺血时,全身免疫功能会受到抑制,导致感染 等并发症发生。但机制不清楚。施教授这项研究显示在缺血性卒中患者和遭受大脑中动脉闭塞小鼠,在大脑和外周自然杀伤(NK)细胞显示不同的活性特性。儿茶酚胺能和下丘脑 - 垂体 - 肾上腺轴的活化通过调节的SOCS3表达导致外周NK细胞数目的减少,而胆碱能神经支配介导的脑中NK细胞应答的抑制。因此,脑缺血通过在脑与外周中不同的机制和通过神经源性神经支配限制中风后感染的靶向抑制来损害NK细胞介导的免疫防御。

秦成峰教授和程根宏教授主要研究zika病毒的免疫机制,以及在zika病毒形成小脑症中的机制, 尤其是25羟基胆固醇作用寨卡病毒的潜在抑制剂。研究发现25羟基胆固醇通过阻断病毒进入 体内抑制ZIKV感染,并且用25羟基胆固醇对小鼠和恒河猴感染zika病毒时提供良好的保护作用, 减少病毒血症,同时还能抑制zika感染诱导小鼠小头畸形。这项研究使得用25羟基胆固醇可能作为天然抗病毒剂打击和预防zika感染及其并发症池洪波教授研究T细胞活化过程中分子网络。
