长非编码RNA(lncRNA)是一些长度超过二百个核苷酸的RNA分子,来自于基因组的非编码区域。虽然lncRNA没有编码任何蛋白质,但它们在不同组织和发育阶段的表达依然具有特异性,这说明lncRNA具有重要的生物学意义。在测序技术的帮助下,人们已经发现了数千种lncRNA,但这些lncRNA的生物学功能依然还是个谜。

人们发现,有些lncRNA通过RNA-蛋白互作招募调控复合物,进而影响附近基因的表达。可能正是因为这个原因,lncRNA表达往往与附近基因的表达相关。不过,这种关联分析起来并不容易。Broad研究所的科学家们最近在这方面取得了重要成果。他们在Nature杂志上发表文章揭示了lncRNA对基因表达的本地调控机制。文章通讯作者是著名遗传学家、美国科学院院士Eric S. Lander教授。Lander教授是Broad研究所的创始人,也是人类基因组计划主要负责人之一。

研究人员对小鼠细胞系进行基因操纵,分析了十二个生产lncRNA的基因组位点。研究显示,其中五个位点顺式影响附近基因的表达。值得注意的是,这些影响不需要lncRNA转录本本身,而是与lncRNA生产过程有关,比如启动子活性、转录过程和转录本剪接。

进一步研究表明,蛋白编码位点也能影响附近基因的表达。六个蛋白编码位点中有四个会对附近基因产生影响。这说明相邻基因的交流是一种普遍现象,涉及多种机制和顺式调控信号。
