PNAS:控制金黄色葡萄球菌致病力的代谢传感器

中国科学院上海药物研究所蓝乐夫研究组、杨财广研究组与复旦大学、芝加哥大学、北卡科他大学等单位的研究人员合作,发现金黄色葡萄球菌利用转录调节因子CcpE来感受自身体内的柠檬酸水平并进而协调自身的代谢状态以及多种致病相关因子的表达,从而实现对细菌致病性的有效控制。相关研究论文Metabolic sensor governing bacterial virulence in Staphylococcus aureus于11月3日以长篇论文形式(Plus)在线发表于《美国国家科学院院刊》(PNAS)。这项研究工作得到了国家自然科学基金委、科技部、上海市科委、中科院“百人计划”等基金的资助。

三羧酸循环是生物机体获取能量的主要方式,也是糖、脂肪和蛋白质三种主要有机物在生命体内分解的共同代谢途径及相互转变的代谢枢纽,而柠檬酸是三羧酸循环中的第一个产物。在该研究中,研究人员发现柠檬酸可与金黄色葡萄球菌的CcpE蛋白结合、促使CcpE蛋白的四聚化并激活CcpE蛋白的转录调节活性;在CcpE蛋白感受、应答柠檬酸信号分子的过程中,第145位、256位精氨酸残基(位于截短型CcpE蛋白上潜在的辅因子结合位点)发挥着重要的作用。

CcpE蛋白不仅可直接激活顺乌头酸酶(三羧酸循环中的酶,催化柠檬酸转变为异柠檬酸)基因的表达从而降低体内柠檬酸的积累,而且还可直接控制编码金黄色葡萄球菌的毒力因子如超抗原样蛋白、荚膜多糖等基因的表达。CcpE的调节子(Regulon)由至少126个基因组成,占金黄色葡萄球菌基因总数目的4.7%以上。有意思的是,虽然CcpE是调节三羧酸循环活性的重要的转录调节因子,在它的调节子中,相当大的一部分基因(40%)编码已知或潜在的金黄色葡萄球菌致病相关因子。

该研究还发现,CcpE通过激活顺乌头酸酶基因的表达从而抑制了金黄色葡萄球菌重要致病相关因子如铁载体、金黄色色素的生成。相应地,CcpE基因的敲除增强了金黄色葡萄球菌在低铁环境下的生长以及对人体血液杀伤作用的抵抗能力。

在小鼠感染模型中,CcpE基因的缺失引起金黄色葡萄球菌在肾脏、肝脏的定植能力显著增强,进一步确定了该基因在金黄色葡萄球菌感染过程中的重要生物学功能。这些研究结果表明CcpE是决定金黄色葡萄球菌致病力的重要的负调节因子。

这项研究首次定义了CcpE蛋白是一个感受柠檬酸的代谢传感器(Metabolic sensor)。同时,该研究发现柠檬酸是控制金黄色葡萄球菌致病性的重要的内源性信号分子,揭示了金黄色葡萄球菌的代谢状态和它的致病性密不可分,为治疗金黄色葡萄球菌感染的替代途径—抗致病力治疗(Anti-virulence therapies)提供了新的理论指导。
