CLIN CANCER RES:阿司匹林通过靶向乙酰肝素酶抑制肿瘤转移和血管生成

近期的流行病学和临床研究表明阿司匹林对肿瘤患者有益,这一发现使得对阿司匹林抗肿瘤能力及其分子机制的研究逐渐受到重视。但是阿司匹林抗肿瘤活性及其相关的机制仍有很多未知。CLIN CANCER RES近期发表了一篇文章,研究对阿司匹林抗肿瘤活性有作用的潜在靶点。

近期的流行病学和临床研究表明阿司匹林对肿瘤患者有益,这一发现使得对阿司匹林抗肿瘤能力及其分子机制的研究逐渐受到重视。但是阿司匹林抗肿瘤活性及其相关的机制仍有很多未知。CLIN CANCER RES近期发表了一篇文章,研究对阿司匹林抗肿瘤活性有作用的潜在靶点。作者使用HTRF技术分析阿司匹林对乙酰肝素酶的影响。使用SPR、分子对接等技术确认乙酰肝素酶是阿司匹林结合蛋白。使用鼠主动脉环模型和鸡胚尿囊膜模型评估阿司匹林抗肿瘤转移和抗血管生成效果,并在B16F10转移模型、MDA-MB-231转移模型和MDA-MB-435PDX模型中进行验证。研究结果表明,乙酰肝素酶,作为一种参与肿瘤转移和血管生成的肿瘤细胞外基质酶,是阿司匹林的潜在靶点。阿司匹林可以直接与乙酰肝素酶Glu225区域相结合抑制酶活性。阿司匹林可以抑制肿瘤转移、血管生成和生长。文章最后认为,该研究结果第一次证明乙酰肝素酶是阿司匹林的靶点。这一发现有助于进一步理解阿司匹林抗肿瘤的机制并为以后乙酰肝素酶小分子抑制剂的发展提供方向。原始出处:Xiaoyang Dai,Juan Yan,et al.Aspirin Inhibits Ca
