国际学术期刊Journal of Biological Chemistry 近日在线发表了生化与细胞所宋建国研究组题为“Hepatocyte Nuclear Factor 6 suppresses the migration and invasive growth of lung cancer cells through p53 and the inhibition of epithelial-mesenchymal transition”的研究论文，该文报道了肝细胞核因子-6在调控上皮-间质转化中的功能及其作用机制。【原文下载】

已知上皮-间质之间转变的可塑性是上皮性肿瘤实现组织浸润和远距离转移的一种重要方式途径。世界卫生组织统计结果显示，上皮癌占全部癌症类型的90%以上，并且是全球范围内导致死亡的一个主要原因。研究上皮-间质之间转化的可塑性发生的分子机制对于有效地控制癌症的恶性转移有着重要的理论指导意义。

宋建国研究组的袁新旺博士等人的最新研究结果显示，在TGF-b1诱导人肺癌细胞发生上皮-间质转化（EMT）的过程中伴随着肝细胞核因子-6（HNF6）表达水平早期发生的和持续性的显著下调。HNF6在肺癌细胞中的表达与细胞上皮表型呈正相关，与间质表型呈负相关。敲减HNF6即可导致肺癌细胞发生EMT，同时增强细胞的迁移能力。而过表达HNF6则能增强肺癌细胞的上皮性，并且抑制细胞迁移，减弱TGF-b1诱导的EMT，抑制细胞的克隆形成、裸鼠皮下肿瘤形成以及癌细胞的浸润能力。进一步的研究表明HNF6主要是通过直接激活抑癌基因p53的启动子来参与调控EMT及细胞迁移。此外，研究显示HNF6在人肺癌组织中的表达与上皮标志性蛋白E-cadherin及抑癌基因p53的蛋白呈高度的正相关性。上述结果表明，HNF6是人肺癌细胞EMT及癌症发展的重要抑制因子。研究成果有助于进一步认识肺癌细胞的可塑性调节及浸润转移相关的重大问题。

该研究工作得到国家科技部，国家基金委的经费资助。
