研究衰老的科学家们将表观遗传学作为衰老过程的原因已经有一段时间了。表观遗传学研究遗传信息和染色体的变化,这些变化不改变基因本身的序列,但确实影响其活性。一种可能性是在我们的细胞中包装DNA从而控制对DNA的访问的组蛋白发生变化。论文通讯作者、马克斯-普朗克老化生物学研究所的Peter Tessarz及其团队如今研究了间充质干细胞的表观基因组。这些干细胞存在于骨髓中,可以产生不同类型的细胞,如软骨细胞、骨细胞和脂肪细胞。

论文第一作者、马克斯-普朗克老化生物学研究所的Andromachi Pouikli解释道,“我们想知道为什么随着年龄的增长,这些干细胞产生的用于骨骼发育和维护的物质越来越少,导致越来越多的脂肪在骨髓中堆积。为了做到这一点,我们比较了年轻和年老小鼠间充质干细胞的表观基因组。我们可以看到,这些干细胞的表观基因组随着年龄的增长而发生明显变化。对骨骼生产有重要作用的基因尤其受到影响。”

间充质干细胞表观基因组恢复青春

这些作者随后调查了间充质干细胞的表观基因组是否能够恢复青春。为了做到这一点,他们用含有乙酸钠的营养物溶液处理从小鼠骨髓中分离出的间充质干细胞。这些干细胞将乙酸钠转化为一种构成单元(building block),酶可以将这种构成单元附着在组蛋白上,以增加对基因的访问,从而提高其活性。Pouikli说,“这种处理令人印象深刻地引起了这些干细胞表观基因组的年轻化,改善了这些干细胞的活性,从而导致骨细胞的产量提高。”

为了阐明这种表观基因组变化是否也可能是人类老年时骨折或骨质疏松症风险增加的原因,这些作者研究了髋关节手术后患者的人类间充质干细胞。来自同样患有骨质疏松症的老年患者的间充质干细胞显示出与之前在小鼠身上观察到的相同的表观遗传学变化。

针对骨质疏松症的新治疗方法?

Tessarz解释道,“乙酸钠(醋酸钠)也可作为食品添加剂,然而,不建议以这种形式使用它来对抗骨质疏松症,因为我们观察到的效果对某些细胞是非常特异的。然而,已经有了干细胞治疗骨质疏松症的初步经验。在这种情况下,用乙酸盐进行治疗也可能起作用。然而,我们仍然需要更详细地调查对整个机体的影响,以排除可能的风险和副作用。”
