最近,研究人员发现,在野生型,但不是pejvakin缺乏(Pjvk - / - ),的听觉毛细胞中,声音过度刺激诱导早期和快速选择性自噬降解过氧化物酶体(pexophagy)。噪声过度暴露引发自噬体相关蛋白MAP1LC3BLC3B;微管相关蛋白1轻链)在野生型而非Pjvk - / - 小鼠中的过氧化物酶体中募集。

研究人员还表明,pejvakin-LC3B结合涉及pejvakin预测的伴侣结构域内的LC3相互作用区域。在转染的细胞和体内转导的听觉毛细胞中,半胱氨酸诱变实验表明,对于活性氧物种诱导的pejvakin-LC3B相互作用和pexophagyC328C343(最接近pejvakinC末端的两个半胱氨酸残基)都需要。用PjvkLc3b cDNA在体内对Pjvk - / - 小鼠的听觉毛细胞进行病毒转导,完全恢复声音诱导的过氧化物酶体自噬,完全阻止氧化应激的发展,并导致过氧化物酶体增殖正常,而Pjvk cDNA单独转导只是部分纠正了缺陷。

总的来说,该研究结果表明,pexophagy在噪音诱导的过氧化物酶体增殖中发挥关键作用,并确定缺陷性的pexophagy是导致噪音诱发的听力损失的原因。
