盘点:抗氧化剂:利大于弊or弊大于利?

抗氧化剂有益的相关研究

【1】脑震荡的潜在疗法——抗氧化剂疗法

近日,来自西弗吉尼亚大学(West Virginia University)的研究人员通过研究表明,抗氧化剂或许在脑震荡的长期效应上扮演着重要作用,而且其或许可以为开发脑震荡的新型疗法提供帮助。

脑震荡在运动员和士兵中较为常见,据估计在美国每年有大约340万的脑震荡患者,而口服补充剂的开发或许可以潜在改善一定比例脑震荡患者的脑部功能;脑震荡往往会引发慢性的损伤性脑病,脑部损伤通常会导致慢性损伤性脑病,这种疾病通常伴随着长期脑损伤及行为症状,比如记忆缺失、冲动行为、抑郁以及侵犯行为等。

Lucke-Wold博士表示,脑震荡会引发脑部功能的长期改变,而这些改变是细胞死亡的主要表现,其很有可能是氧化性压力所引发的,本文研究则显示诸如硫辛酸样的抗氧化剂,当其给予脑震荡患者时会减少患者长期的脑部功能缺失。

【2】抗氧化剂治疗高血压

相关报道

高血压影响超过7000万美国人,是中风,心脏衰竭等肾脏和心血管疾病的主要危险因素。近日,休斯顿大学研究人员大学正在研究抗氧化途径在缓解高血压的作用。

药理学教授Mustafa F. Lokhandwala正在研究肾激素受体,其负责尿钠排泄,被称为排钠的条件,维持血浆钠成分和调节血液压力。

肾脏在调节血容量,钠平衡,pH值和血压中起到关键作用。肾多巴胺受体的激活是控制钠平衡,且随后维持正常血压的一个重要因素。氧化应激,高血压的独立的危险因素,可能会破坏肾多巴胺受体的功能。其结果是,多巴胺不能够促进钠排泄,因为多巴胺的受体不能正常工作,这导致钠潴留和高血压。

肾多巴胺机制任何功能不全会导致过多的钠重吸收,体积膨胀,最终高血压,Lokhandwala说:引起血压升高有各种因素,其中之一是氧化应激。当人体内活性氧或自由基水平增加, 会所造成肾脏荷尔蒙系统损坏,使得易感于高血压等病症。

【3】Gut:富含抗氧化剂食品或可防治胰腺癌

科学界一直在探寻日常饮食与癌症的关联。一项最新研究就显示,常吃富含天然抗氧化剂硒和维生素C、E的食物,或许有助于降低人们患胰腺癌的风险。

英国东英吉利大学等机构研究人员在新一期《内脏》(Gut)杂志上发表报告说,他们对2万多人进行了跟踪研究,期间这些受调查者需要汇报他们的日常食物种类、分量以及采用何种烹饪方式等。

分析显示,那些通过食物摄入硒等抗氧化剂物质最多的受调查者,与摄入这类营养物质最少的那部分人相比,患胰腺癌的风险要低67%。这类营养物质常存在于谷类、坚果、水果和蔬菜等食物中。

【4】Biol Psych:新型抗氧化剂或可抑制精神分裂症和抑郁症的发展

来自瑞士洛桑大学的研究者对引发精神分裂症的原因和引发氧化性压力的易感基因之间的关联性关系进行了数年的研究,氧化性压力可以归咎于大脑发育期间的感染、炎症、哮喘或者心理社会压力,这就意味着在儿童期以及青春期处于风险状态的项目尤其容易暴露,但是一旦个体进入成年期就不会受到影响了。

研究者的相关研究刊登于国际杂志Biological Psychiatry上,该项研究通过对谷胱甘肽缺失的小鼠进行研究,谷胱甘肽是一种保护细胞免于氧化性压力的重要分子,其可以使得神经元更多地暴露于氧化压力所引发的有害作用下。研究者发现,当小鼠处于年轻期时,如果其受到压力,则大脑中的微清蛋白神经元就会受损,而且这种损伤是终生存在的,更有意思的是,对于成年鼠来说给予相同的压力,却对成年鼠的微清蛋白神经元没有任何影响。

【5】ARS:抗氧化剂或成为潜在的帕金森疾病疗法

近日,来自乔治亚健康科学大学的研究者的最新研究报道,一种新型强大的抗氧化剂有一天将有可能成为治疗帕金森疾病的潜在药物。这种抗氧化剂为合成的三萜类化合物,可以阻止帕金森疾病的发展。相关研究成果刊登在了近日的国际杂志Antioxidants & Redox Signaling上。

研究者Thomas和其同事使用抗氧化剂药物来支持Nrf2,从而可以阻止帕金森病人脑细胞中所产生的多巴胺的消失。Nrf2是氧化压力和炎症的主要调节子,可以明显降低早期的帕金森患者疾病进程,实际上Nrf2的活性随着年龄逐渐下降。在帕金森患者中,你可以看到其氧化性应激的负担,这就是为什么研究者选择这个靶点了。

【6】JAD:抗氧化剂可能对阿尔茨海默氏症有效

一项发表在《阿尔茨海默氏病杂志》上的新的研究证实:抗氧化剂可能有助治疗阿尔茨海默氏症。

阿尔茨海默氏病(AD)是美国人死亡的第六大原因,超过65岁近八分之一的人受阿尔茨海默氏病影响。目前还没有治疗手段能治愈这种疾病。然而越来越多的证据表明改变身体处理铁和其他金属如铜和锌的方式可能在AD症状开始出现之前就已经发生了变化。一项新的研究表明降低血浆中的铁的含量可能保护AD患者的脑。

在医学及健康科学学院北达科他州大学药理学及生理学副教授Othman Ghribi博士领导的研究小组开展的这项研究中,研究人员给兔子喂食高胆固醇的饮食,使兔子脑部积累β-淀粉样蛋白(Aβ)的一个小蛋白斑块。这些斑块具有神经元毒性,对阿尔茨海默氏病的发生发展至关重要。兔子tau蛋白发生了变化,tau蛋白是神经细胞骨架的一部分。当这种蛋白质被过度磷酸化后,神经细胞之间用电信号进行交流的能力被削弱。兔子在给予去铁酮(一种铁螯合剂)后,血浆中的铁含量减少,大脑中的β-淀粉样蛋白和tau蛋白的磷酸化水平也恢复到正常水平。

抗氧化剂有害的相关研究

【7】Science子刊:含抗氧化剂的糖尿病药物或可促进癌症发生转移

近日,一项刊登在国际杂志Science Translational Medicine上的研究论文中,来自中国第三军医大学等机构的研究人员通过对小鼠研究发现,糖尿病药物中的抗氧化剂或可促进癌症扩散;研究者指出,某些糖尿病药物或可刺激肿瘤的转移,包括结肠癌和肝癌等,而本文研究结果一旦在人类机体中被证实,那么糖尿病患者就应当注意包含抗氧化剂药物的使用了。

目前大量动物研究都揭示了抗氧化剂的潜在用途,抗氧化剂可以保护细胞免于毒性活性氧自由基的损伤,从而就会加速癌细胞的生长和扩散,通常抗氧化剂用于治疗糖尿病患者;研究者认为糖尿病患者通常患许多癌症的风险均较高,而且患癌症的糖尿病患者的数量也与日俱增,然而糖尿病药物到底如何影响癌症的发生和发展,目前研究者并不清楚。

【8】STM:人造抗氧化剂加速小鼠皮肤癌转移

近日,来自瑞典的科学家在国际学术期刊science translational medicine上发表了一项最新研究进展,他们指出一种用于帮助慢性阻塞性肺病(COPD)病人祛痰的人造抗氧化剂药物--N-乙酰半胱氨酸可能会加速小鼠皮肤癌的扩散,这一研究结果对该药物的安全性提出了新的质疑。

该药物除了用于帮助COPD病人祛痰,还有一些人将其用作保健品,他们认为抗氧化剂可以减少运动相关肌肉损伤,还可以燃脂以及降低疲劳。

研究人员用掺有N-乙酰半胱氨酸的水对肿瘤小鼠模型进行处理,发现虽然这种抗氧化剂对皮肤肿瘤的数目和大小没有明显影响,但会加速黑色素瘤细胞向身体其他部位的迁移和侵袭。他们还发现接受N-乙酰半胱氨酸处理的小鼠淋巴肿瘤的数目会加倍,之前研究也已经表明特定的抗氧化剂还会加速小鼠肺部肿瘤的生长。

【9】Cell Reports:大量抗氧化剂恐会对神经干细胞有害

根据赫尔辛基大学的分子神经学研究项目中的Anu Wartiovaara研究小组的一项研究表明,干细胞对氧自由基和抗氧化剂尤为敏感。该项研究由研究人员Riikka Martikainen发表在2015年5月28日《Cell Reports》上。

线粒体是细胞能量工厂,它可使用氧气产生能量。在该过程中可产生一种副产品也就是活性氧。过多的氧自由基会引起细胞损伤,但少量的氧自由基是必须的,因为它们是一种重要的胞信号分子。氧自由基的一个主要功能是控制干细胞的功能。抗氧化剂广泛用于阻止活性氧的作用,因为活性氧会对身体造成一定的损害。为了增强抗氧化剂的作用效果,一些新的抗氧化剂会针对性的积累到线粒体中。

【10】抗氧化补充剂:不宜过多

“维他命C是抗氧化剂。抗氧化剂对您有益。喝下吧,您会觉得好多了。”广告商很聪明,把话说的如此动听,让人觉得似乎真是那么回事。但是别忘了:科学上没有100%确定无疑的事。认识量是认识的深化,抗氧化补充剂虽好,多则不宜。

受补充剂供应商的鼓吹,公众似乎一直误认为自由基对身体有害,而抗氧化物则为佳品。当然,就像影响我们健康的众多其他因素一样,事情远非那么简单。

自由基是含有未配对电子的分子或原子。未配对电子处处惹事生非。它们生性不喜落单,因而一直在寻找其它电子,也因此异常活跃。自由基就像那个醉醺醺、跌跌撞撞来到派对,然后开始搞砸一切的损友。抗氧化剂分子则能够捐出一个电子给自由基,把自由基稳定下来。它们就像耐心的诤友,能够劝服自由基不要酒后驾车,而后领进客房,让他醒醒酒,万一吐了,还给端茶递桶。

【11】Nature研究再证明食用抗氧化剂会促进癌症转移

来自美国德州大学西南医学中心儿童研究所的研究人员进行了一项研究,他们发现相比于正常细胞,癌细胞从抗氧化物得到的获益更多,这一发现增加了人们对于癌症病人食用饮食中抗氧化物的担心。(生物谷之前曾报道抗氧化剂会加速小鼠皮肤癌细胞转移。)

癌细胞转移是癌细胞从原发部位传播到身体其他部分的过程,是导致多数癌症病人死亡的重要原因。该研究团队发现对癌症小鼠模型进行抗氧化物处理会使癌细胞扩散更快。相关研究结果发表在国际学术期刊Nature上。

长久以来人们已经知道癌细胞从一个部位向身体其他部位传播是一个非常低效的过程,绝大多数进入血液中的癌细胞最终都不能存活。

【12】NEJM:抗氧化剂如何加速癌症发生

近几十年来,世界各地追求健康的人们食用抗氧化剂补品和富含抗氧化剂的食物,他们认为这是一个健康和长寿之路。

然而关于抗氧化剂补品的临床试验一再打破消费者的希望即抗氧化剂能减少患癌症的风险。几乎所有的相关实验都没有表现出任何它能抗癌的作用。事实上,在一些试验中抗氧化剂补品与特定癌症的发生率增加相关联。在一项试验中,吸烟者吸取额外的β-胡萝卜素会患有更高风险的肺癌发生率。

刊登在《新英格兰医学杂志》上的一条简论提出为什么抗氧化补充剂可能不会降低癌症发展,以及为什么他们可能实际上弊大于利。
