PNAS：抵御耐药性细菌感染的重大突破

近日一项刊登在杂志PNAS上的研究论文中，来自斯坦福大学等处的科学家们通过研究鉴别出了调节细菌毒素毒力的细胞组分，尤其是这种名为PLEKHA7的蛋白质，通过消除这种蛋白的表达，细胞就会从α-毒素的伤害中恢复过来，缺失PLEKHA7蛋白的小鼠会送细菌感染中恢复过来，同时也会增强肺炎的生存率。

金黄色葡萄球菌强大的侵袭力取决于α-毒素，该毒素可以通过刺穿细胞膜来破坏细胞，而研究者发现，该菌的粘着连接（adherens junctions）在组织感染的发生及感染的扩散上扮演着重要角色，Lauren Popov说道，我们的研究表明，粘着连接的很多组分都会参与控制α-毒素的毒力，目前我们推测蛋白PLEKHA7扮演着主要的角色。

为了确定PLEKHA7在调节金黄色葡萄球菌毒力中的重要性，研究者感染了不能表达编码PLEKHA7蛋白的细胞，结果发现，这些细胞从α-毒素的损伤中获得了恢复的能力，而且通过利用缺失PLEKHA7蛋白的多重耐药的金黄色葡萄球菌感染小鼠，研究者发现，小鼠可以从感染中快速恢复过来，同时也增强了小鼠患肺炎的生存率。

目前研究者正在尽力研究理解PLEKHA7蛋白如何控制α-毒素的活性，其中一种假设就是，该蛋白可以通过传输信号诱导细胞自我毁灭来聚集细菌的毒素，而当PLEKHA7控制机体皮肤感染和致死性肺炎的严重性时，研究者就可以通过靶向作用这种粘着连接过程中的非必要组分，使其作为潜在靶点来降低多重耐药性细菌的毒力。

最后研究者指出，阐明在细菌感染期间PLEKHA7蛋白的重要性对于后期开发治疗细菌性感染，尤其是耐药性细菌感染的新型疗法或将带来一定帮助和希望。
