近日,国际杂志 《Clinical Chemistry》上在线发表一项关于肺癌治疗监测结果中干血滴中的微小RNA的技术稳定性和生物学变异性的研究。

多项研究将miRNA用作血液生物标志物。但是特别值得注意的是,从患者收集血液的方法可以显著影响诊断结果。

研究人员探讨干血滴(DBS)是否促有利于对miRNA的稳定测量,并将其技术稳定性与生物变异性进行比较。首先,研究人员通过对暴露于不同温度和湿度条件的下的来自1个人18个全基因组范围的miRNA谱产生DBS样品来测试其稳定性。其次,从1人再次进行7次重复DBS技术重复。最后,研究人员调查了53肺癌患者进行不同疗法的DBS样本。在这3个阶段中,产生了来自DBS108个全基因组miRNA谱,并以生物统计学方法对其进行了评估。

在稳定性分析中,研究人员观察到温度和湿度对miRNomes的影响总体有限(不同条件下的平均相关性为0.993)。硅胶的使用略微降低了DBS的技术重现性。7项技术重复平均相关系数为0.996。与同一人全血PAXGene miRNomes的相关性显著(相关性为0.88)。最后,对53例暴露于不同疗法的肺癌患者的样本进行评估,结果显示生物学变异显著超过技术变异(P <0.0001),产生51个变化的miRNA

以从DBS收集样品为基础,研究人员提供了稳定的整个miRNomes的分析工作流程。研究发现生物差异显著超过技术变异。基于DBSmiRNA谱可能进一步深化对miRNA生物标志物翻译特征的研究。
