研究人员将Cu2 +纳米粒子添加到阴离子羧甲基壳聚糖(CMC)和海藻酸盐(Alg)的混合物中,而不是直接向聚合物混合物中添加抗菌Cu2 +离子导致不受控制的聚合物交联。然后,从Cu2 +离子Cu2纳米粒子中释放逐渐与聚合物混合物交联,通过冷冻干燥进一步转化为具有互连多孔结构的支架(CMC/Alg/Cu)。

结果显示,与通过相同程序制造的CMC/Alg支架相比,CMC/Alg/Cu支架可明显改善支架的成骨能力和杀死细菌的能力。具体而言,体外研究表明,具有良好生物相容性的CMC/Alg/Cu支架可以通过上调粘着相关基因(粘着斑激酶(FAK),桩蛋白(PXN)和纽蛋白(VCL))的表达水平来增强前成骨细胞的粘附,通过上调成骨相关基因表达和细胞外钙沉积来促进成骨分化和矿化。体内研究进一步表明,CMC/Alg/Cu支架可在4周内诱导血管化新骨组织的形成,同时避免了临床细菌感染,甚至当临床收集的金黄色葡萄球菌感染种植部位时。

综上所述,该研究结果表明,本研究开发了一种简单新颖的制造铜聚合物支架的方法,所制备的支架或可用于修复感染的骨缺损。
