近日,来自日本的科学家们在国际期刊nature medicine上发表了他们的最新研究进展,他们发现DNA甲基转移酶3a(DNMT3a)在调节骨代谢与骨细胞分化方面具有重要作用。
 
研究人员指出,当细胞所处环境发生变化,细胞会进行代谢重组以进行应答,进而调节细胞分化过程,但联系代谢过程与分化过程的具体分子机制仍不清楚。在这项研究中,研究人员发现DNA甲基转移酶3a能够作为转录因子将破骨细胞的代谢变化与分化过程联系起来。同时,在破骨细胞分化过程中,细胞的代谢水平会向氧化代谢过程偏移,而对破骨细胞的生成具有重要作用的细胞因子RANKL能够诱导向氧化代谢过程偏移的代谢变化,还伴随着SAM的合成增加。
 
研究人员发现SAM介导的DNMT3A对DNA的甲基化能够通过表观遗传抑制抗破骨细胞生成基因的表达调节破骨细胞产生。DNMT3A缺失的破骨前体细胞不能正常分化为破骨细胞,同时,DNMT3A在破骨细胞特异性缺失会导致小鼠骨密度增加,这些发现均说明DNMT3A在骨代谢中具有非常重要的作用。在骨质疏松模型中进一步发现,通过抑制DNA甲基化能够有效减少骨质疏松。
 
综上所述,这项研究揭示了表观遗传在细胞代谢与分化过程之间的调控作用,或许可为治疗骨相关疾病提供新的治疗策略。
