【1】PNAS:老药新用治疗乳腺癌

来自美国莱斯大学的研究人员近日通过研究发现了一种新方法,该方法能够抵御和乳腺癌增殖相关的特殊蛋白的过度表达,研究者指出,降低NAF-1蛋白的水平以及其运输的铁硫簇分子(iron-sulfur clusters)的活性或许就能够帮助抑制肿瘤生长,相关研究刊登于国际杂志PNAS上。

研究者认为,用于治疗2型糖尿病的药物-吡格列酮或许就能够有效控制NAF-1蛋白的水平,NAF-1的单突变几乎会完全阻断癌细胞增殖的能力,这或许就支撑了研究者的一种看法,即降低NAF-1的水平就能够帮助阻断肿瘤。调节特异性应对肿瘤的药物或许就能够为科学家们提供一种强大的武器来帮助抵御乳腺癌或其它癌症;然而NAF-1蛋白的过度表达或许就和前列腺癌、胃癌、宫颈癌、肝癌及喉癌直接相关。(文章详见--PNAS:老药新用治疗乳腺癌

【2】Nature子刊:乳腺癌还与细菌有关?

最近,科学家发现女性乳腺组织中也有共生菌群,并且这些菌群与乳腺癌有关。这项研究由美国梅奥诊所(Mayo Cllinic)的科学家们完成,并发表在了《Scientific Reports》上。

研究人员发现,从无菌手术室中取出的病人乳腺组织中存在着细菌DNA,而病人并无细菌感染的症状。这些菌群与覆盖乳腺的皮肤上的菌群完全不同,更重要的是,乳腺癌患者乳腺组织中的菌群与健康人乳腺组织中的菌群有显著的差别,并且良性乳腺肿瘤患者和恶性乳腺肿瘤患者组织中的菌群也不尽相同。恶性乳腺癌患者带有大量的梭杆菌属细菌,这些细菌可能会释放炎症因子造成局部炎症反应,从而促进癌细胞的产生。(文章详见--Nature子刊:乳腺癌还与细菌有关?

【3】JAMA Oncol:导管上皮非典型增生后的乳腺癌风险研究

导管上皮非典型增生(ADH)是乳腺癌的危险因素。研究者进行了一项研究,比较不同诊断方式(空芯针穿刺活检 vs 切除活检)下的ADH与浸润性乳腺癌风险之间的联系。

队列研究比较了955 331名进行钼靶筛查女性的浸润性乳腺癌10年累积风险(诊断或未诊断ADH)。数据来自于5个乳腺影像学注册数据库。其中1727例ADH患者,空芯针穿刺活检和切除活检分别诊断了1058 (61.3%)例和635 (36.8%) ADH患者。平均诊断年龄为52.6 (46.9-60.4)岁。

【4】Lancet Oncol:乳腺癌新标志物:SPAG5

增殖标志物已被推荐用于指导乳腺癌系统治疗的选择。不过最好的标志物是什么事还不知道。本研究旨在确认乳腺癌的增殖驱动因素及其相关特征,评估这些标志物与临床结果、化疗反应间的联系。

研究数据来自3个队列:诺丁汉发现队列(n=171),乌普萨拉队列(n=249),国际乳腺癌联盟的分子分类学(METABRIC)队列(n=1980)。对精子相关抗原5(SPAG5)进行了重点分析,调查了SPAG5 基因拷贝数变化、mRNA转录表达、蛋白质表达的临床病理相关性;分析了SPAG5 基因拷贝数变化、mRNA转录表达、蛋白质表达等与乳腺癌特异性生存率、无病生存率、无远处转移生存率、病理完全缓解和残余癌症负荷等的关系。(文章详见--Lancet Oncol:乳腺癌新标志物:SPAG5

【5】Lancet Oncol:乳腺癌和肺癌患者全身抗癌治疗后30天死亡率研究

基于全身抗肿瘤治疗(SACT)数据集,评估英国接受SACT的乳腺癌和肺癌患者的30天死亡率。

这项基于人口的研究,纳入了女性乳腺癌患者,以及肺癌患者(男性和女性),大于等于24岁,在2014年开始SACT,不管先前的治疗周期或治疗方式,也不管目前的疾病情况。我们计算了这些患者最近一次SACT周期后的30天死亡率。校正相关混杂因素,使用Logistic回归分析,探究患者、肿瘤或治疗相关因素是否与30天死亡率有关。

【6】CA:年轻的乳腺癌患者,你检测了BRCA基因吗?

最近的一项研究表明,年轻乳腺癌患者接受BRCA突变基因检测的比例在提高。目前美国国家综合癌症网络(NCCN)指南推荐所有≤45岁乳腺癌女性进行BRCA检测,即使在已知其他风险因素的情况下。

本项多中心前瞻性队列研究纳入了1126名≤40岁的乳腺癌患者。大多数的参与者(85%)教育程度在大学或以上,99%的人有保险。约87%的女性报告说他们在诊断乳腺癌、调查后1年内检测了BRCA基因突变;并且这一比例从2007年的70%增加到2012年的96%,再到2013年的95%。(文章详见--CA:年轻的乳腺癌患者,你检测了BRCA基因吗?

【7】PLoS One:对乳腺癌的诊断排除,MRI提供100%筛查

来自生物医学成像与图像引导治疗部门的维也纳医科大学的研究者们对来自14个国际研究组的超过2300名的患者进行了元分析。分析首次发现,如果乳腺癌筛查结果不明确,那么在后续程序中进行MRI检查,将能够百分百排除对癌症的筛查。“如果MRI扫描不显示肿瘤,则进一步其它的疾病危险也将被排除在外。一旦不存在疾病担心的恐惧,那么受影响的女性则可以安然入睡了,”分子生物学家和放射学家Barbara Bennani-Baiti和放射学家Pascal Baltzer表示。(文章详见--PLoS One:对乳腺癌的诊断排除,MRI提供100%筛查

【8】PLOS MED:超过14万的女性数据表明,基因预测BMI值越高,乳腺癌风险越低

观察性流行病学研究表明,对于绝经前妇女,较高的BMI与较低的乳腺癌风险有关;但是对于绝经后妇女,较高的BMI却与较高的乳腺癌风险有关。这些联系是否是通过共享遗传或环境因素所介导的呢?

我们使用了两项大型乳腺癌数据们进行孟德尔随机分析,评估BMI和乳腺癌风险间的关系。我们利用84个BMI相关遗传变异,建立了加权BMI遗传评分,用于预测BMI。使用BCAC中的个体水平数据,评估了基因预测BMI与乳腺癌风险之间的联系(46,325例患者和42,482例对照);接着使用DRIVE项目中的汇总数据,再次对基因预测BMI与乳腺癌风险之间的联系进行了评估。由于大多数研究是在诊断后才测量BMI,我们不能进行平行分析,以充分评估BMI和乳腺癌风险。

结果显示,通过全基因组关联研究(GWAS)识别的预测BMI的相关遗传变异,与绝经前或绝经后的乳腺癌风险呈负相关。在这项研究中,观察到遗传预测BMI的增加,与绝经后乳腺癌风险的降低有关,这个结果不同于以前BMI与乳腺癌之间的阳性关联。为什么会这样,还需要进一步的机制研究。(文章详见--PLOS MED:超过14万的女性数据表明,基因预测BMI值越高,乳腺癌风险越低
