Science:阿尔茨海默病新突破:在发现β-淀粉样蛋白100多年后,科学家终于确定其高清结构!

1901年,德国医生Alois Alzheimer接诊了一位51岁的老妇人。她患有严重的记忆障碍,讲话困难并且难以理解别人话,对时间与地点也毫无概念。Alzheimer医生意识到这是一种前所未知的疾病。在病人去世后,Alzheimer医生对其进行尸检,惊奇地发现患者的大脑中有许多淀粉样蛋白斑和神经纤维缠结。

这就是人类至今仍束手无策的阿尔茨海默症(Alzheimer disease,AD)最主要的两大病理特征:错误折叠的β-淀粉样蛋白(Aβ)在大脑中沉积形成的淀粉样蛋白斑块和Tau蛋白过度磷酸化导致的神经纤维缠结。一个世纪后,科学家们终于确认了这两种AD生物标志物的高清结构。今年7月5日, MRC Laboratory of Molecular Biology的研究人员曾在Nature上通过冷冻电镜(cryo-EM)揭示了Tau蛋白的高分辨率蛋白结构。两个月后,德国和荷兰的一组研究人员同样利用冷冻电镜确定了另一种AD相关蛋白——Aβ的高清结构。这一发现再次为AD药物的研发带来了新的希望。

1.Aβ的高清结构

在这项发表于9月7日Science期刊上的研究中,来自德国尤里希研究所、杜塞尔多夫大学和马斯特里赫特大学等机构的科学家们阐明了Aβ原子层面的三维结构:许多单个Aβ层层交错排列成所谓的原丝(protofilaments)。两个原丝相互缠绕,形成一个原纤维(fibril)。如果几个这样的原纤维缠结在一起,那么这就形成了在AD患者脑组织中的典型沉积或斑块。这些结构细节可以解答许多关于有害沉积物生长方面的问题,同时也解释了遗传风险因素的影响。

“在对淀粉样蛋白结构和相关疾病的基础理解上,这是一个里程碑,”该研究的通讯作者Dieter Willbold教授解释道,“原纤维的结构解答了许多和纤维生长机制有关的问题,并确定了一系列导致早发AD的家族基因突变的作用。”

这个高清结构的分辨率达4Å,即0.4纳米,属于原子半径和原子键长度的量级。与之前的研究相比,该模型首次展示了蛋白质的确切位置和相互作用。缠绕的原丝中的Aβ分子并没有在同一水平,而是像拉链一样交错间隔。此外,结构首次阐明多个Aβ蛋白质分子中全部的42个氨基酸残基的位置和构象。

这一新颖、详细的结构为理解一些增加患病风险的基因修饰的结构效应提供了新的基础。它们通过改变某些位点的蛋白质的模板来稳定原纤维。这也解释了为什么在自然界中,小鼠并不会患上阿尔茨海默氏症,为何一些人群对改变的易感性存在差异。

2.多种方法的综合应用

与100多年前Alois Alzheimer医生发现的蛋白斑不同,新研究揭示的原纤维结构无法通过光学显微镜观察——而是利用低温电子显微镜技术。科学家们在马斯特里赫特大学花了一年多的时间来分析冷冻电镜的数据。此外,使用固态核磁共振(NMR)光谱学和x射线衍射也有助于进一步补充和支持原纤维结构的图像,并验证所获得的数据。

“低温电子显微镜下的单个图像通常噪声很大,因为蛋白质对电子辐射非常敏感,这些图像只能在非常低的辐射强度下产生。”研究人员解释道。他们使用计算机辅助程序,将数千张不同的图像结合起来,从中提取出高分辨率的结构数据。

“如果样本是异质的,也就是说由不同的结构的原纤维组成,那么这是一个非常复杂的步骤。这是过去淀粉样蛋白的常见情况,也是分析的主要障碍之一。然而,我们现在有非常均一的原纤维样品——90%的原纤维都具有相同的形状和对称性。”研究通讯作者之一Schroder说。

来自尤里希研究所的Lothar Gremer博士成功地生成了原纤维样本。“其中关键的一步是大大延迟样本中原纤维的生长速度——从几小时到几周。因而每个Aβ分子有足够的时间以一种统一和高度有序的方式排列成均一的原纤维。”Gremer补充道。

固体核磁共振波谱学的研究提供了额外的数据来建立模型并帮助验证结构。“NMR使我们能够获得更多的信息,比如哪个氨基酸残基形成了盐桥,从而提高了原纤维的稳定性,” 研究人员之一Henrike Heise教授解释道。由汉堡结构系统生物学中心的Jorg Labahn教授主持的x射线衍射实验进一步证实了这一结果。
