本研究作者应用系统生物学的方法检查抑制分子的特征,提出它们可以有助于斑块稳定性假说。

研究者将人类颈动脉斑块(n = 177)与健康动脉(n = 15)作微阵列分析发现,相比健康动脉组,颈动脉斑块组肌肉收缩、肌肉发育和肌动蛋白细胞骨架这几条信号通路下调最多。除了典型的平滑肌细胞标记物之外,这些途径中还包括以前与动脉粥样硬化无关的细胞骨架相关基因。SYNPO2,SYNM,LMOD1,PDLIM7和PLN基因的表达与斑块中典型的平滑肌细胞标记物和大鼠内膜增生呈正相关。免疫组化发现蛋白表达在正常血管的平滑肌细胞,在人斑块和内膜增生中缺如。大多数蛋白质定位于平滑肌细胞的细胞骨架,染色质免疫沉淀发现这一过程可被组蛋白的化学修饰活性增强剂H3K27ac调节。机制方面,血小板衍生生长因子β、干扰素γ、暴露于剪切流应力和氧化低密度脂蛋白超载均可抑制这些基因表达。PDLIM7,PLN和SYNPO2基因变异与没有心血管疾病症状的高风险受试者(n = 3378)的颈动脉内膜中层厚度的进展相关。通过eQTL分析,rs11746443也与斑块中的PDLIM7表达相关,体外沉默PDLIM7导致平滑肌细胞标记物表达下调、肌动蛋白细胞骨架被破坏、细胞迁移减少和增殖能力增加。

本研究确定一组反映血管疾病平滑肌细胞表型改变的基因,它们可能是平滑肌细胞去分化的早期敏感标志物。
