Nat Neurosci:唐焰等揭示“社会抚摸”背后的神经生物学机制

中国文化里向来认为拥抱和抚摸是过于亲密的行为而羞于谈及,而在西方社会看来却是极为普通自然的社交行为。心理学上认为拥抱和抚摸能快速拉近人与人之间的距离、消除陌生感、增加亲密感情并保持身心健康。

是什么冲动让相互陌生的心灵间产生交流的渴望?是何种力量让万水千山难以阻挡亲友间的思念?曾高谈阔论,然知己难觅;金迷纸醉只得黄粱一梦;脉脉而视君不见... 孰不知《月下独酌》中的李白或许仅仅是友人默默地轻柔地一次抚摸,便能消去他邀月对饮影零乱的寂寞惆怅……

中国文化里向来认为拥抱和抚摸是过于亲密的行为而羞于谈及,而在西方社会看来却是极为普通自然的社交行为。心理学上认为拥抱和抚摸能快速拉近人与人之间的距离、消除陌生感、增加亲密感情并保持身心健康。这其中尤为重要的一个假说就是拥抱和抚摸触发了一种神经肽:催产素(Oxytocin, OT)的释放而让人身心愉悦并产生信任感,然而多年来因为缺乏技术手段一直难以从神经科学角度证明。另外也归咎于催产素并非字面意义上那么简单,作为五亿年进化过程中横跨脊椎动物门中各个纲目物种的神经肽,它有着调节哺乳生产、性行为、社交、信任、伴侣、母性以及抗焦虑、恐惧和疼痛等多重功能,同时OT神经元深藏于下丘脑的两个小核团(室旁核及视上核),在体研究其放电活动是一项挑战。

为了探寻这个问题,2020年7月27日,德国海德堡大学Valery Grinevich和斯特拉斯堡大学Alexandre Charlet合作(共同一作为唐焰博士、Diego Benusiglio、Arthur Lefevre和Louis Hilfiger)在Nature Neuroscience杂志上发表文章Social touch promotes interfemale communication via activation of parvocellular oxytocin neurons,首次揭示了催产素系统与社交抚摸之间的正反馈机制。研究人员选择社交活动丰富的雌性大鼠作为实验对象,避免了雄鼠间攻击行为与雌雄间性行为的实验干扰。通过特异的光遗传学标记结合在体微电极记录,首先发现背部肢体接触确实快速触发催产素能OT神经元的同步放电激增,且嗅觉视觉和听觉刺激并无显着影响。

其次在结构与功能上,研究人员分别用在体光纤钙成像、微透析与离体膜片钳验证了由小细胞控制大细胞的OT神经元微环路。接着更深入探究了两种类型OT神经元在全脑的不同输入来源,证明小细胞OT比大细胞OT有更多的传入突触并有独特脑区的输入源。最后研究人员又通过遗传药理学操控少数小细胞OT神经元,成功激活或抑制全体OT神经元活性从而实现了人工调控雌性大鼠间的社交行为,增加或减少肢体接触时间。

社交过程中肢体接触有效诱发OT神经元放电增加 在麻醉状态下,背部吹气airpuff能有效再现OT神经元放电增加。光纤成像fiberphotometry结合DREADD特异调控体感刺激airpuff对OT神经元的响应,左侧红色表示药物激活小细胞后体群体OT对airpuff更为敏感;右侧则是抑制小细胞后群体OT对airpuff无反应。绿色为对照组OT神经元对airpuff反应。

这项研究开拓了体感刺激介导内源性神经肽释放的先河,并在神经环路上找到特异调控靶点小细胞OT神经元;为开发特异而高效的促神经肽释放药物提供了理论基础,尤其在自闭症和创伤后应激综合症等精神疾病的治疗上带来新的希望。

背部体感刺激从脑干传入下丘脑小细胞OT神经元,后者同步激活大细胞OT神经元,释放催产素至中枢及外周神经系统从而增加社交动机。这一研究或许第一次用催产素解释了“海内存知己,天涯若比邻”的神经机制,同时也提醒各位在后新冠时代中社交隔离会降低催产素释放而造成精神创伤,请务必进行干预及心理治疗。无法拥抱握手,拍背可否成为健康又安全的新社交礼仪。
