辉瑞酪氨酸激酶抑制剂Inlyta治疗早期肾癌的临床三期试验惨遭失败

一项ATLAS临床III期试验评估了Inlyta作为肾切除术后复发性肾细胞癌(RCC)高危患者辅助治疗的潜力。独立的数据监测委员会进行分析后发现,以延长无病生存期(DFS)为终点,与安慰剂组相比,Inlyta治疗组患者并没有明显改善。

Inlyta是辉瑞公司开发的新一代口服酪氨酸激酶抑制剂(TKI),选择性的作用于血管内皮生长因子1、2、3受体(VEGFR),抑制血管生成、肿瘤生长和癌症的进展。该药物在2012年1月17日获得美国食品药品监督管理局(FDA)批准用于治疗对其他药物无应答的晚期肾癌患者。

辉瑞全球产品开发部肿瘤学首席发展官Mace Rothenberg表示:"我们对这项研究的结果感到很失望,因为我们曾经希望Inlyta作为晚期肾癌患者的二线治疗药物的可以扩展到治疗早期肾癌患者,达到延缓或者预防疾病的复发,可是这个目标并没有实现。接下来我们会对数据做额外的分析,以便更深刻的了解这一结果。Inlyta联合免疫检查点抑制剂治疗RCC在内的多种晚期癌症的研究将继续进行。
