J Endod:二甲双胍介导AMPK通路增强牙髓细胞向成牙本质细胞的分化

二甲双胍是治疗II型糖尿病的一线药物,它可以调节间充质干细胞的分化。然而,它对人牙髓细胞(DPCs)的作用还未可知。这篇研究的目的是为了调查二甲双胍对DPCs增殖和分化的作用。研究使用活/死活力测定试剂盒检测二甲双胍对DPCs细胞活力的影响。使用细胞计数试剂盒(CCK-8; Dojindo, Tokyo, Japan)检测细胞增殖情况。在施加二甲双胍和腺苷5'-单磷酸酯活化的蛋白激酶(AMPK)信号抑制剂Compound C (EMD Chemicals, San Diego, CA)的情况下,通过western blot检测AMPK的磷酸化及非磷酸化水平。通过碱性磷酸酶活性和von Kossa染色检测Compound C对二甲双胍诱导的DPCs的成牙本质细胞分化的作用,并通过反转录聚合酶链式反应检测成牙本质分化标志物的表达变化。结果显示,流式细胞仪检测结果表明,DPCs展现出间充质干细胞的特性。不同剂量的二甲双胍均展现出与DPCs良好的相容性,细胞活力>90%。高达50μmol/ L的二甲双胍浓度不会影响细胞增殖。Western blot结果显示,DPCs表达功能性的有机阳离子
