通过迈克尔型加成反应,由含二丙烯酸酯的聚乙二醇基支架和半胱氨酸封端的肽交联剂(CGPQG↓IWGQC)合成水凝胶。因为CGPQG↓IWGQC可被基质金属蛋白酶8(MMP-8)切割,盐酸米诺环素,牛血清白蛋白或抗菌肽(KSL)被掺入支架中以按需给药系统的MMP-8响应释放行为。使用凝胶时间,傅里叶变换红外光谱,扫描电子显微镜和流变参数测量检查水凝胶表征和凝胶动力学。通过高效液相色谱和蛋白质特异性测定进行降解行为和MMP-8响应性药物释放。生物相容性评价表明水凝胶是无细胞毒性的。抗菌测试表明释放的药物能够维持生物活性。

总之,这些结果表明MMP-8敏感性水凝胶是按需口内局部药物递送的有希望的候选者。由于MMP-8是牙周炎最重要的生物标志物之一,MMP-8反应性水凝胶有可能用于慢性牙周炎和种植体周围炎的原位适应性降解。这个概念必须在牙周病的动物模型中进行测试。
