Cell:重大突破!rigosertib被证实是抑制RAS癌基因的小分子

RAS基因(HRAS、KRAS和NRAS)突变经常在很多最为常见和致命的肿瘤(包括胰腺癌、肺癌和结肠癌)中观察到。尽管分子肿瘤学家在理解这些突变和它们对细胞信号转导的影响方面取得重大进展,但是在开发系统性地靶向这些RAS癌基因的药物方面几乎没有取得任何进展。这种进展的缺乏导致这个领域的很多人将RAS视为一种“无药可靶向(undruggable,也译作无药可治的,无药可用的)”的癌基因。

在这项研究中,研究人员鉴定出首个小分子能够同时抑制RAS癌基因激活的不同信号通路。这个被称作rigosertib或ON01910.Na的小分子作为一种蛋白-蛋白相互作用抑制剂发挥作用,阻止RAS与将正常细胞变成癌细胞的信号蛋白(包括RAF和PI3K等)相结合。研究人员开展结构学实验,证实了rigosertib的这个作用机制,并且证实这种靶向作用机制有潜力用于治疗几种由RAS癌基因驱动的癌症。

Reddy博士说,“这一发现是癌症领域的一个重大突破。rigosertib的作用机制代表着一种攻击无药可靶向的RAS癌基因的新策略。我们当前的重点是使用来自我们利用rigosertib开展的研究中提供的信息,设计下一代小分子RAS靶向疗法,而且激动人心的是,我们最近鉴定出我们认为可改善rigosertib性质的几种化合物。”

在包括西奈山伊坎医学院在内的几个地方,这种候选药物当前处于III期临床试验中用于治疗骨髓增生异常综合征(myelodysplastic syndrome, MDS)。


在此前rigosertib与gemcitabine 联合用于转移性胰腺癌的研究中,已进入临床II/III期。Rigosertib (ON 01910.Na)作为 Ras mimetic和PLK1、PI3K的小分子抑制剂使用。

2013年,在加州大学旧金山分校细胞和分子药理学系主任、霍华德休斯医学研究所Kevan Shokat博士领导下,该研究小组设计出了一些小分子,它们可以不可逆转地结合到ras蛋白一种从前未知的、短暂存在的口袋内,当突变ras蛋白改变形状之时这一口袋才会显露出来。他们的研究工作在线发表在11月20日的《自然》(Nature)杂志上。(Nature:靶向K-ras突变的小分子化合物研发成功 攻克30年挑战

2015年,斯坦福大学医学院的研究人员Siprashvili Z发现在12种常见人类癌症,包括皮肤癌、乳腺癌、卵巢癌、肝癌和肺癌中,10-40%的肿瘤缺失一对称作为SNORD50A/B的snoRNAs。他们指出肿瘤缺失SNORD50A/B的乳腺癌患者,和相比正常组织,肿瘤中这些RNAs分子水平降低的皮肤癌患者,比同类患者存活下来的可能性更小。Khavari说:“我们搜查了在癌细胞中高度异常的一些基因组区域。惊讶地发现SNORD50A/B在如此多不同类型的癌症中频繁缺失。它们缺失的频率与其他一些非常著名的肿瘤抑制基因一样。”Khavari 说:“令人震惊地是,我们发现这些RNA结合了RAS家族中的一些蛋白,尤其是KRAS。这真是我们期待的最后一件事。其特别令人惊讶是因为十多年来我的实验室一直在对KRAS开展深入研究,因此这真是太巧了。”(Nat Genet:一种管家型snoRNAs分子是著名癌分子K-RAS的开关)
