本研究旨在探讨临床宫颈标本中HPV 16长控制区(LCR) E2BSs的甲基化状态。

研究人员采用焦磷酸测序法定量检测43例临床HPV 16感染宫颈标本中4个E2BSs的甲基化状态。同时，使用Quantivirus®HPV E6/E7 RNA 3.0 assay (bDNA)检测相应标本中E6/E7 mRNA水平。

研究结果显示，晚期鳞状上皮内病变（HSIL）和宫颈癌中E2BS1,2和4位点的甲基化状态显著高于无症状HPV16感染和低度鳞状上皮内病变（LSIL）（所有P <.05）。此外，HPV 16 E2BS1和2的甲基化状态与E6 / E7 mRNA水平正相关（分别为rs = 0.529和0.512，P <.01）。接受者操作特征曲线分析用于比较E2BSs甲基化的诊断性能。当Youden指数为最大值时，E2BS1和E2BS2的甲基化水平均显示出最佳诊断灵敏度为77.8％，特异性分别为80％和92％。

研究表明，E2BS1和2的甲基化状态可以作为未来宫颈病变严重程度的诊断指标。
