Diabetologia:环境污染与糖尿病的关系

短暂暴露于TCDD可导致胰岛中CYP1A1酶活性的长期升高。这为胰岛在体内直接暴露于亲脂污染物提供了证据,并可能对胰岛生理产生影响。

一直认为,环境污染与人类糖尿病发病率有关,但潜在的致病机制仍不清楚。考虑到调节胰岛素分泌在维持葡萄糖稳态中的关键作用,到达内分泌胰腺并引起细胞损伤的环境化学物质尤其值得关注。我们认为,细胞色素P450 (CYP)酶参与代谢外源性生物,可以作为一个有用的生物标志物直接暴露于污染物的胰岛。此外,胰岛内的功能性CYP酶也会影响细胞生理。本研究的目的是确定CYP1A酶在直接或系统暴露于环境污染物后是否被激活。

用已知的CYP1A诱导剂2,37,8-四氯二苯并对二恶英(TCDD)和3-甲基蒽醌(3-MC)体外处理永生化肝脏(HepG2)和啮齿动物胰腺内分泌细胞系(MIN6βTC-6INS1αTC1αTC3)以及人胰岛。此外,小鼠在体内注射单个高剂量TCDD或多个低剂量TCDD,并在1714天后分离胰岛。

研究结果表明,体内多次低剂量TCDD暴露也导致小鼠胰岛Cyp1a1显著上调。体外直接暴露于人和小鼠胰岛可抑制葡萄糖诱导的胰岛素分泌。单次注射大剂量TCDD可降低血浆胰岛素水平,并显著增加细胞死亡。
