最近,一项刊登于国际杂志Proceedings of the National Academy of Sciences上的研究论文中,来自约翰霍普金斯大学的研究人员通过对小鼠进行研究发现,癌细胞很少凭自己形成转移性肿瘤,而是喜欢在机体个组织间“巡游”最终成功地完成转移;同时研究者还发现,移动中的细胞同原发性肿瘤部位中进行繁殖的细胞并不相同,而这种区别就可以增加移动肿瘤细胞对化疗的耐受性。

研究者Andrew Ewald说道,癌细胞通常会增加形成新型转移病灶的机会,其会开启分子程序来帮助癌细胞在机体多样性环境中移动。在第一批实验中,研究者Ewald及其团队对小鼠进行工程化操作使其在特殊的光下显现红光或绿光,随后移除小鼠机体中的肿瘤细胞,并且在小鼠的乳腺中植入发红光和绿光的肿瘤,这些细胞并不会发光,其会在肺部中寻找转移点。

如果单一细胞移居到新的转移点,细胞聚集物就会发出一种光,而研究者却发现了许多发出多种颜色光的肿瘤细胞聚集物,利用计算机模型进行研究他们发现,大部分单一色彩的聚集物都来自多种细胞,这些细胞都会发射出一种颜色的光,研究者估计转移性癌症中低于3%的细胞来自于单一的细胞。

随后研究者对比了单一细胞和细胞簇形成细胞克隆的能力,首先他们对装有明胶样商业产品(可以模拟乳房肿瘤周围的环境)的培养皿中两组相同数量的细胞进行检测,结果发现,相对于单一肿瘤细胞而言,肿瘤细胞簇形成单克隆的能力是前者的15倍,当在小鼠机体中重复实验后,这些细胞簇产生大量转移性肿瘤的能力就变成了单一肿瘤细胞的100倍。

研究者Ewald说道,我们想观察是否移动中的细胞可以表现出特殊的分子特征,从而被我们用来预测并且开发抑制肿瘤扩散的方法,此前实验室研究结果表明,从肿瘤组织中脱颖而出的细胞通常会被名为K14的细胞所修饰;而K14的水平在原发性肿瘤和大型转移性肿瘤中非常低,但却在小型移动的细胞群簇中非常高;培养皿实验结果表明,细胞可以在两种分子程序间开启,即在细胞增殖和转移间开启,而K14的水平则可以解释给定细胞的特殊程序。

由于化疗药物可以靶向作用增殖中的细胞,而转移性的细胞则不会被这种药物所杀灭,从而使得癌症患者对新型肿瘤更加易感,本文研究发现或可帮助患者克服这种易感性;如今研究者可以利用蛋白质K14来分离两种类型的细胞,同时还以对比两种细胞的基因活性,研究者还希望这项研究可以用来开发新型药物对转移性的肿瘤细胞进行靶向作用。
