据《中国科学报》记者了解,长期服用抗氧化剂的功效尚没有明确的研究证据,相反已有研究证据表明,抗氧化剂能促进肿瘤的生长和转移,并不适合肿瘤病人。而鲁林荣课题组的最新发现,是第一次提示了抗氧化剂对于自身性免疫疾病的风险。

鲁林荣教授长期从事免疫调节和自身免疫性疾病的研究,特别关注一类叫做T淋巴细胞的免疫细胞。其中Th17细胞是科学家在2005年新发现的一种辅助性T细胞亚群。Th17细胞在维持肠道稳态、抵抗胞外细菌、真菌感染的宿主防御中起到重要作用。

“如果手指上划破一个伤口,外界的细菌、真菌就会入侵人体,为了,为了抵御这些入侵或感染,人体的免疫系统会被激活,通过炎症反应来将入侵者‘清理’出去。伤口从出现到愈合,免疫反应会经过启动、高潮又回到原点的调控过程。”鲁林荣说,“在正常情况下,包括Th17细胞在内的各种辅助性T细胞的数量和活性维持着动态平衡,如果某一类细胞太多了,机体就会产生免疫反应失衡,导致免疫反应低下或是反应过度,损伤到正常的体细胞。”大多自身免疫疾病与免疫反应过度有关。常见的自身免疫系统疾病包括:多发性硬化、红斑狼疮、风湿性关节炎、痛风等等。

鲁林荣课题组研究发现,一种激酶MINK1能通过感知细胞内或是细胞微环境中的活性氧水平抑制Th17的分化。如果把Th17比作是战斗力极强的莽夫,那MINK1就是劝莽夫要小心行事的警察,帮助维持机体的免疫平衡。

他们的研究还发现,但在某些特定的情况下,这种平衡调节会被打破,引发和促进免疫反应过度,那就是抗氧化剂的摄入。在小鼠试验中,给小鼠喂食一种抗氧化剂——N-乙酰半胱氨酸(NAC)后,会诱发小鼠产生更严重的自身免疫性脑炎。第一作者博士生付国通说,这相当于在抗氧化剂面前,MINK1对于TH17的抑制减弱了,使Th17细胞更加容易“反应过度。”

“这个研究提示我们,服用抗氧化剂存在风险,即破坏了我们人体的免疫平衡,特别是针对那些对自身免疫性疾病易感的人群。”鲁林荣说。

我们生活中常见的抗氧化剂有NAC、维生素C、辅酶Q-10,SOD,GSSG(氧化型谷胱甘肽)等等,它们可以中和细胞内的自由基,因此被商家拿来作为保健品或化妆品的成分,并植入能够“延缓衰老,永葆青春”的概念。“但事实上,学术界对于抗氧化剂的作用一直存在争议,目前并没有确切的实验表明额外服用抗氧化剂对身体有利,因此必须慎重采用。”
