Science:吕奔教授团队揭示重度中暑诱发脏器损伤的关键机理

该团队近年来还揭示了危重症DIC与多脏器衰竭形成的重要机理。

5月5日,中南大学湘雅三医院吕奔教授团队在国际学术期刊Science上在线发表了题为“Z-DNA binding protein 1 promotes heatstroke-induced cell death”的研究论文,揭示了热射病(又称“重度中暑”)的重要致死机理——高体温通过ZBP1蛋白诱发过度的程序性细胞死亡,进而导致危及生命的弥散性血管内凝血(DIC)与多脏器损伤。该论文的第一单位为中南大学湘雅三医院。中南大学博士后袁芳芳为第一作者,吕奔教授为通讯作者。

高体温诱发的危重症为热射病。但高体温如何导致脏器损伤与机体死亡的机制过去不甚清楚。吕奔教授团队利用前期脓毒症研究中获得的线索,进一步揭示了高体温通过诱发ZBP1依赖的程序性细胞死亡在热射病发生发展中引起的致死效应,颠覆了既往的学术观点“高体温通过物理性损伤导致脏器功能衰竭”,展示了温度感应、程序性细胞死亡、DIC与脏器功能衰竭之间的内在联系,为热射病等危重症的防治提供了重要思路。

2021年的诺贝尔生理学或医学奖表彰了“温度和触觉感受器的发现”,凸显了朱利叶斯团队的重要发现——细胞膜上的辣椒素受体TRPV1是一种能被高温活化的离子通道。吕奔教授团队的这项研究明确提示了细胞浆内也存在“温度感受器”,受到高温刺激时能诱发ZBP1依赖的程序性细胞死亡。该生命现象虽然可能在抗感染免疫中发挥保护作用,但在持续性高体温作用下可导致DIC、多脏器损伤甚至死亡。

此外,该团队近年来还揭示了危重症DIC与多脏器衰竭形成的重要机理,以中南大学为第一单位发表了系列NI指数论文,包括Science、Immunity、Blood、Nat Commun和J Biol Chem,得到了学术界的高度评价。
