TSC蛋白复合物位于胰岛素/生长素（IIS）信号转导通路中的中心节点，通过GAP蛋白活性来抑制mTOR复合物1 (mTORC1) 的活性，进而调控细胞生长。TSC复合物由三个蛋白质组成，即TSC1/TSC2以及近来鉴定的TBC1D7。其中TSC1/TSC2在人类中突变会导致多发性肿瘤；而TBC1D7在人类中突变可导致巨头症（Macrocephaly），但其致病机理尚不清楚，有待研究。

中国科学院遗传与发育生物学研究所降雨强研究组与北京生命科学研究所王涛实验室合作，运用果蝇遗传学以及生物影像学等技术手段对TBC1D7的生理功能进行研究，发现TBC1D7在果蝇中完全突变会导致个体变大的表型。该表型是一种非细胞自主性的个体系统性生长，主要是由突变体内胰岛素类似多肽2（ILP2）的合成与分泌增加所致。研究揭示了TBC1D7对ILP2合成与分泌的调控不依赖于传统的TSC复合物的功能。

该研究首次报道了TBC1D7的不同于传统TSC蛋白复合物的生理功能，为研究TBC1D7突变与人类疾病的关系提供了重要依据。相关的研究结果已经在线发表于Journal of Cell Biology 。该项目得到了国家自然科学基金项目与”973”项目的支持。
