Br J Cancer:非靶向代谢组学研究揭示与乳腺癌发病风险相关的代谢物

乳腺癌作为女性中最常见的癌症之一,约占全球所有女性癌症病例的25%。

乳腺癌作为女性中最常见的癌症之一,约占全球所有女性癌症病例的25%。既往研究显示,饮酒、肥胖以及出生时体重和身高的增加会提高该疾病的患病风险,而体育锻炼、母乳喂养以及钙的摄入量也具关联性。

了解诊断前相关代谢作用可以潜在地帮助识别风险更高的人群,并更好的了解疾病的早期发生机制。先前的少数代谢组学相关研究探索了乳腺癌发病风险与血浆或血清代谢物浓度之间的关联性。该研究旨在获得更详细的研究来了解和预防该疾病。

研究人员通过基于核磁共振(NMR)的非靶向代谢组学方法,对来自791个乳腺癌病例和791个来自E3N(EPIC-France)队列的匹配对照的基线血浆样品进行了分析。通过NMR谱建立PLS-DA(偏最小二乘判别分析)模型,以预测疾病的结果,并通过条件逻辑回归分析43种已鉴定的代谢物的比值比和FDR(错误发现率)调整的CI(置信区间)。

参与者及代谢物数据的描述性分析

结果显示,绝经前亚组的乳腺癌发病率预测具有一定的准确性,且鉴定了与疾病发病风险相关的10种代谢物,包括组氨酸、N-乙酰糖蛋白、甘油以及乙醇。但在总体人群或绝经后妇女均未发现显著的预测疾病的相关代谢物。

总而言之,在绝经前而非绝经后的乳腺癌患者中观察到相关的代谢紊乱。其中组氨酸和NAC与炎症发生相关,而乙醇与发病风险之间的显著关联性也暗示了酒精摄入或与乳腺癌的发病相关。
